中文摘要: 为研究提取方法对海带多糖的空间结构与生物活性的影响,以海带干粉为原料,通过热水、酸辅助、碱辅助提取,得到3种海带多糖(polysaccharides from Laminaria japonica,LJP)。通过高效液相色谱(high performance liquid chromatography,HPLC)法、刚果红试验等方法来研究海带多糖的空间结构特征,探究海带多糖的空间结构与其流变性质之间的关联,并且建立细胞氧化损伤模型进一步研究海带多糖对细胞氧化损伤的保护作用。结果表明,碱辅助提取海带多糖(polysaccharides from Laminaria japonica by base-assisted,BLJP)具有较高的产率([9.52±0.05)%],酸辅助提取海带多糖(polysaccharides from Laminaria japonica by acid-assisted,ALJP)的糖醛酸含量最高([37.56±0.88)%],热水提取海带多糖(polysaccharides from Laminaria japonica by hot water,HLJP)的硫酸基含量最高([15.96±0.44)%];HLJP、ALJP、BLJP的重均分子量分别为1 547.94、104.10、2 547.65 kDa;剪切流变曲线表明海带多糖溶液是具有剪切稀化现象的非牛顿流体,Power-Law线性拟合表明BLJP的稠度系数最高,为1.275 1 Pa·sn;ALJP的稠度系数最低,为0.207 9 Pa·sn,与其重均分子量相对应;ALJP浓度5 mg/mL时,其羟基自由基清除率最高,为81.08%,可以使H2O2氧化损伤的RAW264.7细胞的丙二醛(malondialdehyde,MDA)含量由2.53 nmol/mL降低至1.57 nmol/mL。
Abstract:To investigate the effect of extraction methods on the spatial structure and biological activity of polysaccharides from Laminaria japonica (LJP),three types of LJP were obtained from Laminaria japonica dry powder by hot water,acid-assisted,and base-assisted extraction. High performance liquid chromatography(HPLC),Congo red reagent,and other methods were used to study the spatial structure characteristics of LJP,explore the correlation between the spatial structure and rheological properties of LJP,and establish an oxidative damage model of cells to further investigate the protective effect of LJP on oxidative damage of cells. The results showed that LJP by base-assisted (BLJP) extraction had a high yield [(9.52±0.05%)],while LJP by acid-assisted (ALJP) extraction had the highest content of glucuronic acid [(37.56±0.88%)]. LJP by hot water(HLJP) extraction had the highest content of sulfate groups [(15.96±0.44%)];their weight average molecular weights were 1 547.94 kDa(HLJP),104.10 kDa(ALJP),and 2 547.65 kDa(BLJP),respectively;the shear rheological curve indicated that the LJP solution was a non-Newtonian fluid with shear thinning phenomenon. Power-Law linear fitting showed that the highest viscosity coefficient of BLJP was 1.275 1 Pa·sn,and the lowest viscosity coefficient of ALJP was 0.207 9 Pa·sn,corresponding to its weight average molecular weight;At 5 mg/mL,the highest hydroxyl radical scavenging activity of ALJP was 81.08%,which could reduce the secretion of malondialdehyde (MDA) in RAW264.7 cells damaged by H2O2 oxidation from 2.53 nmol/mL to 1.57 nmol/mL.
