中文摘要: 为探究紫苏叶花色苷的抗氧化活性及模拟胃肠道消化对其活性的影响,该研究通过pH示差法测定紫苏叶花色苷在体外模拟口腔和胃肠道消化前后花色苷的含量变化,并进行DPPH、ABTS+、羟自由基抗氧化活性测定。结果表明:消化前紫苏叶花色苷DPPH、ABTS+、羟自由基最大的清除率分别为79.51%、86.92%、85.75%,DPPH、ABTS+、羟自由基清除率与花色苷浓度成正比,紫苏叶花色苷抗氧化活性优于VC组。经模拟胃消化3.0 h后,不同浓度的试验组花色苷释放量均为最大值,对照组却无明显变化。在模拟肠道消化过程中,pH值的变化导致花色苷含量呈下降趋势。消化1.0 h后DPPH、ABTS+、羟自由基清除率均达到最大值,分别为92.73%、95.36%、95.30%。
Abstract:To investigate the antioxidant activity of anthocyanins from Perilla frutescens leaves and the effect of simulated gastrointestinal digestion on their activity,this study measured the changes in anthocyanin content before and after in vitro simulated oral and gastrointestinal digestion using the pH differential method,and evaluated the antioxidant activity by DPPH,ABTS+,and OH radical scavenging assays. The results showed that before digestion,the maximum scavenging rates of DPPH,ABTS+,and OH radicals by P. frutescens leaf anthocyanins were 79.51%,86.92%,and 85.75%,respectively,with the scavenging rates positively correlated with anthocyanin concentration. The antioxidant activity of P. frutescens leaf anthocyanins was superior to that of the vitamin C (VC) group. After simulated gastric digestion for 3.0 h,the anthocyanin release reached the maximum at all experimental concentrations,while no significant changes were observed in the control group. During simulated intestinal digestion,changes in pH led to a downward trend in anthocyanin content. After digestion for 1.0 h,the scavenging rates of DPPH,ABTS+,and OH radicals all reached their maximum values,at 92.73%,95.36%,and 95.30%,respectively.
