中文摘要: 以铁皮石斛为研究对象,采用不同剂量的60Co-γ 射线辐照一次性处理(1、2、5、10、15、25 kGy)和相同剂量的2次辐照处理,对比辐照前后铁皮石斛中菌落总数、粗多糖、总黄酮、甘露糖等物质的含量及其抗氧化活性等的变化,确定铁皮石斛辐照灭菌的安全剂量。结果表明,10 kGy以上的辐照剂量能够将铁皮石斛中的微生物完全杀死,随着贮藏时间延长,辐照残留量逐渐降低,90 d后,辐照后的铁皮石斛光释光(photostimulated luminescence,PSL)值筛查结果为阴性。10、15 kGy辐照剂量下,铁皮石斛中的粗多糖、总黄酮、甘露糖等含量及其乙醇提取物的DPPH自由基清除能力未受到影响,与空白样品相比,各类指标含量及体外活性均有增加;但经25 kGy剂量的一次辐照或经10、15、25 kGy剂量2次辐照后,与一次辐照的铁皮石斛相比,其功效成分含量降低,其乙醇提取物的DPPH自由基清除能力下降。综合各项指标变化情况,铁皮石斛的60Co-γ 射线辐照安全剂量为10 kGy,且功效成分含量及抗氧化活性增加,相同剂量的2次辐照或高剂量辐照会降低铁皮石斛的功效成分含量及其抗氧化活性,但其蛋白质含量没有明显变化,这可能与蛋白质检测方法有关。

Abstract:This study took Dendrobium officinale as the research object and used different doses of 60Co-γ ray irradiation for one-time treatment(1,2,5,10,15 kGy,and 25 kGy)and two irradiation treatments at the same dose.The aerobic plate count,crude polysaccharides,total flavonoids,mannose,and other substances in Dendrobium officinale before and after irradiation,as well as their DPPH free radical scavenging ability,were compared to determine the safe dose for irradiation sterilization of Dendrobium officinale.The results showed that a radiation dose above 10 kGy could completely kill the microorganisms in Dendrobium officinale.With prolonged storage time,the residual radiation gradually decreased,and after 90 days,the photostimulated luminescence(PSL)value screening result of Dendrobium officinale after irradiation was negative.At radiation doses of 10 kGy and 15 kGy,the contents of crude polysaccharides,total flavonoids,mannose,and the DPPH free radical scavenging ability of the ethanol extract in Dendrobium officinale were not affected.Compared with the blank sample,the contents and in vitro activity of various indicators increased.However,after one-time irradiation at a dose of 25 kGy or two irradiations at doses of 10,15 kGy,or 25 kGy,the contents of active components of Dendrobium officinale decreased compared with the first irradiation,and the DPPH free radical scavenging ability of the ethanol extract decreased.Based on the changes in various indicators,the safe dose of 60Co-γ ray irradiation for Dendrobium officinale was 10 kGy,and the contents of active components and antioxidant activity increased.Two irradiations at the same dose or high-dose irradiation reduced the contents of active components and antioxidant activity of Dendrobium officinale,but there was no significant change in its protein content,which may be related to the protein detection method.
