基于HS-SPME-GC-MS结合ROAV技术解析速食骨汤酸菜排骨挥发性风味物质变化

中文摘要: 以酸菜和猪排骨为主要原料,研究贮藏过程中速食骨汤酸菜排骨挥发性风味物质变化。采用最优工艺制备速食骨汤酸菜排骨,利用顶空固相微萃取-气质联用(headspace-solid phase microextraction-gas chromatography-mass spectrometry,HS-SPME-GC-MS)和电子鼻技术,结合相对气味活度值(relative odor activity value,ROAV)和正交偏最小二乘法判别分析(orthogonal partial least squares discrimination analysis,OPLS-DA)法,分析不同贮藏时间挥发性风味物质变化。电子鼻主成分分析表明,0、15 d和25 d靠近X轴,气味特征相似,20 d远离X轴与其他贮藏期差异较大;HS-SPME-GC-MS鉴定出6类52种挥发性风味物质,不同贮藏时间(0、5、10、15、20、25 d和30 d)分别检出32、37、34、35、31、28种和27种挥发性风味物质,其中醛类相对含量高达29.78%、20.53%、20.09%、25.20%、21.73%、19.88% 和20.35%,辛醛、癸醛、已醛和(E,E)-2,4-癸二烯醛风味物质占比较高;酸类相对含量分别为16.04%、34.19%、30.11%、19.55%、38.27%、42.51% 和41.96%,乙酸、棕榈酸和月桂酸风味物质占比较高,与电子鼻分析结果相同;结合ROAV和OPLS-DA,(E,E)-2,4-癸二烯醛、月桂酸、草蒿脑、辛醛、癸醛和二甲基三硫可作为区分不同贮藏时间骨汤酸菜排骨差异的关键风味物质。
Abstract:With sauerkraut and pork ribs as the main ingredients,this study aims to investigate the changes in volatile flavor compounds in instant sauerkraut and pork ribs with bone broth during storage. The optimal process for preparing instant sauerkraut and pork ribs with bone broth was used,and changes in volatile flavor compounds during storage were analyzed by using headspace solid-phase microextraction-gas chromatographymass spectrometry(HS-SPME-GC-MS) and electronic nose technology,combined with relative odor activity value (ROAV) and orthogonal partial least squares discrimination analysis (OPLS-DA). The principal component analysis of the electronic nose showed that the samples after storage of 0,15,and 25 d were close to the Xaxis,with similar odor characteristics,while the sample after storage of 20 d was far from the X-axis and significantly different from the other storage periods. A total of 52 volatile flavor compounds from six categories were identified by HS-SPME-GC-MS. During different storage periods (0,5,10,15,20,25,and 30 d),32,37,34,35,31,28,and 27 volatile flavor compounds were detected,respectively. The aldehyde compounds accounted for high relative contents of 29.78%,20.53%,20.09%,25.20%,21.73%,19.88%,and 20.35%,with a high proportion of octanal,decanal,hexanal,and (E,E)-2,4-decadienal. The acid compounds had relative contents of 16.04%,34.19%,30.11%,19.55%,38.27%,42.51%,and 41.96%,with acetic acid,palmitic acid,and lauric acid being the most prevalent flavor compounds. These results were consistent with the electronic nose analysis. According to ROAV and OPLS-DA analysis,the key flavor compounds for distinguishing the differences between sauerkraut and pork ribs with bone broth during different storage periods were (E,E)-2,4-decadienal,lauric acid,estragole,octanal,decanal,and dimethyl trisulfide.
