中文摘要: 该研究探究长双歧杆菌婴儿亚种B79(Bifidobacterium longum subsp. infantis B79,B79)活菌及其热灭活菌介导免疫途径对3T3-L1前脂肪细胞成脂分化和脂质代谢的影响。以适宜浓度的B79活菌及灭活菌悬液为试验组,以肽聚糖为阳性对照,以RPMI1640培养基为空白对照,分别与RAW264.7细胞共培养后测定细胞因子的表达及分泌量。共培养上清液经离心后以适当的比例干预3T3-L1细胞并测定脂质代谢相关基因的表达情况及脂联素的分泌量;油红染色后倒置显微镜下观察3T3-L1细胞成脂情况并测定油红O萃取值OD490。结果表明,与空白对照组相比,B79活菌及灭活菌干预后,RAW264.7细胞免疫功能被激活,白细胞介素-6(interleukin-6,IL-6)、白细胞介素-10(interleukin-10,IL-10)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-1β(interleukin-1β,IL-1β)的表达及分泌量显著提高。经共培养上清液干预3T3-L1细胞后,试验组成脂基因及脂质积累基因的表达被抑制,脂联素的分泌量降低,其中,B79灭活菌组脂联素的分泌量及油红O萃取值(OD490)最低。长双歧杆菌婴儿亚种B79及其热灭活菌可激活巨噬细胞的免疫调节功能,从而抑制前脂肪细胞的成脂分化和脂质代谢功能;B79灭活菌的效果更为稳定,抑制成脂的能力优于B79活菌。
Abstract:The effects of live and heat-inactivated Bifidobacterium longum subsp. infantis B79 on the immune pathways that mediate adipogenesis and lipid metabolism of 3T3-L1 preadipocytes were studied. The experimental groups were treated with appropriate concentrations of live and heat-inactivated B79 suspensions,with peptidoglycan as the positive control and RPMI1640 medium as the blank control. After co-culture with RAW264.7 cells,the cytokine expression and secretion were measured. The supernatant of the co-culture system was centrifuged and then used to intervene in 3T3-L1 cells at appropriate ratios,and the expression levels of lipid metabolism-related genes and the secretion of adiponectin were determined. After Oil Red staining,the adipogenesis of 3T3-L1 cells was observed under a microscope and the OD490 value of Oil Red O extraction was measured. The results showed that compared with the blank control group,the live and heat-inactivated B79 groups showed significantly activated immune function of RAW264.7 cells and significantly increased expression and secretion of interleukin-6(IL-6),interleukin-10(IL-10),tumor necrosis factor-α(TNF-α),and interleukin-1β(IL-1β). After intervention with the co-culture supernatant,the expression of genes involved in adipogenesis and lipid accumulation in the experimental group was significantly inhibited,and the secretion of adiponectin was significantly reduced. The secretion of adiponectin and the OD490 value were the lowest in the heat-inactivated B79 group. The live and heat-inactivated cells of B. longum subsp. infantis B79 significantly activated the immune regulatory function of macrophages,thereby inhibiting the adipogenesis and lipid metabolism of preadipocytes. Moreover,heat-inactivated B79 demonstrated more stable effect,outperforming live B79 in inhibiting adipogenesis.
