中文摘要: 为探究发酵黑米膳食纤维对非酒精性脂肪性肝病(non-alcoholic fatty liver disease,NAFLD)小鼠中肝脏脂质沉积的影响及作用机制,通过蛋氨酸-胆碱缺乏(methionine and choline deficiency,MCD)饮食构建NAFLD小鼠模型,分别应用低剂量(2.5 g/kg bw)和高剂量(5.0 g/kg bw)发酵黑米膳食纤维对小鼠进行处理。4周后采样检测小鼠肝组织病理学,测定血清学标志物丙氨酸氨基转移酶(alanine aminotransferase,ALT)、天门冬氨酸氨基转移酶(aspartate aminotransferase,AST)水平;定量聚合酶链式反应(quantitative polymerase chain reaction,qPCR)分析肝脏脂质代谢和转运相关酶基因ACACA、FASN、TGR5、CPT1α、FABP1、CD36、APOB、MTTP、FXR的mRNA表达情况。结果表明,低剂量组小鼠肝脏的病理形态略有改善,血清学标志物及脂质代谢和转运相关酶基因的mRNA表达水平改善不明显;高剂量组小鼠肝脏的病理形态改善明显,血清学标志物水平显著降低(p<0.05),脂质代谢和转运相关酶基因的mRNA表达显著上调(p<0.05)。因此,高剂量发酵黑米膳食纤维能够改善NAFLD,可能与其上调脂质代谢相关转运基因的信号途径相关。
Abstract:To investigate the effects and mechanisms of dietary fiber of fermented black rice on hepatic lipid deposition in mice with non-alcoholic fatty liver disease(NAFLD). An NAFLD mouse model was established using a methionine and choline deficiency (MCD) diet. The mice were treated with low-dose (2.5 g/kg bw)and high-dose (5.0 g/kg bw) dietary fiber of fermented black rice. After four weeks, liver histopathology was examined, and serological biomarkers, including alanine aminotransferase(ALT) and aspartate aminotransferase (AST) levels, were measured. Quantitative polymerase chain reaction (qPCR) was used to analyze the mRNA expression of genes related to lipid metabolism and transport, including ACACA, FASN, TGR5,CPT1α, FABP1, CD36, APOB, MTTP, and FXR. The results showed that in the low-dose group, there was a slight improvement in the pathological morphology of the mouse liver, while the changes in serological biomarkers and the mRNA expression levels of enzymes related to lipid metabolism and transport were not significant. In contrast, the high-dose group exhibited a marked improvement in the pathological morphology of the mouse liver, a significant reduction in serological biomarkers (p<0.05), and a significant upregulation in the mRNA expression of enzymes associated with lipid metabolism and transport(p<0.05). Hence, high-dose dietary fiber of fermented black rice could ameliorate NAFLD, which may be associated with its upregulation of signaling pathways related to lipid metabolism and transport genes.
