中文摘要: 该文选用5种不同聚合度的壳寡糖以及纤维二糖、松二糖功能寡糖,由噻唑蓝(methylthiazolyldiphenyl-tetrazolium bromide,MTT)法筛选模型建立所使用的皮质酮(corticosterone,CORT)浓度,并进行浓度筛选,基于乳酸脱氢酶(lactate dehydrogenase,LDH)检测研究功能寡糖对CORT诱导的PC-12细胞的保护能力。利用Annexin V-FITC/PI双染法流式细胞仪对细胞凋亡率进一步检测,并采用蛋白免疫印迹分析测定细胞凋亡相关蛋白进行探究。结果表明,添加壳四糖与纤维二糖能显著降低细胞凋亡率,对CORT诱导的神经元应激损伤模型的保护效果相对较好。
Abstract:Five kinds of chitooligosaccharides with different polymerization degrees and common functional oligosaccharides such as cellobiose and pine biose were selected to study.Firstly,the concentration of corticosterone(CORT)used was established by methylthiazolyldiphenyl-tetrazolium bromide(MTT)screening model,and concentration screening was performed. The protective ability of functional oligosaccharides on CORT-induced PC-12 cells was studied based on lactate dehydrogenase(LDH)detection.Annexin V-FITC/PI double staining flow cytometry was used to further detect the apoptosis rate,and the apoptosis-related proteins were determined by western blot analysis. The experimental results showed that the addition of chitotetracose and cellobiose significantly reduced the apoptosis rate,and had the best protective effect on CORT-induced neuronal stress damage model.
