中文摘要: 抗性淀粉含量影响食品的生理功能,并且直接决定富含抗性淀粉原料的加工特性。准确测定食品中抗性淀粉含量是抗性淀粉应用的前提。影响抗性淀粉含量测定的关键因素是去除样品中的非抗性淀粉和样品中抗性淀粉的水解。该试验选取高直链玉米抗性淀粉(7012)、红豆和红薯为研究对象,通过单因素试验优化去除非抗性淀粉和水解抗性淀粉的最佳条件。去除样品中非抗性淀粉的最佳条件为添加α‐胰腺淀粉酶30 U/mL+淀粉葡萄糖苷酶3 U/mL,于37 ℃水浴振荡水解16 h,50%乙醇溶液洗涤2 次。水解抗性淀粉的最佳条件为添加3 260 U/mL 淀粉葡萄糖苷酶0.10 mL 于50 ℃水浴振荡水解30 min。对建立的方法进行精密度、准确度、检出限和定量限验证,结果显示,样品中抗性淀粉含量10 次测定结果的相对标准偏差小于10%,表明方法精密度好;已知抗性淀粉含量的质控样品测定值与标示值间的误差为2.62%,表明方法准确度高。综上,该文所建立方法适用于食品中抗性淀粉含量的测定。
Abstract:The content of resistant starch affects the physiological functions of food and directly determines the processing characteristics of raw materials rich in resistant starch. Accurately measuring the content of resis‐tant starch in food is a prerequisite for its application. The key factors affecting the determination of resistant starch content are the removal of non‐resistant starch from the sample and the hydrolysis of resistant starch in the sample. The experiment selected high amylose corn resistant starch(7012),red bean,and sweet potato samples as research objects,and optimized the optimal conditions for removing non‐resistant starch and hydro‐lyzing resistant starch through single‐factor experiments. The optimal conditions for removing non‐resistant starch from the sample were determined by adding 30 U/mL α‐amylase and 3 U/mL starch glucosidase and shaking in a water bath at 37 ℃for 16 h of hydrolysis,followed by washing twice with 50% ethanol solution.The optimal conditions for hydrolyzing resistant starch involved adding 0.10 mL of 3 260 U/mL starch glucosi‐dase and shaking in a water bath at 50 ℃for 30 min. The method was validated for precision,accuracy,detec‐tion limit,and quantification limit. The results showed that the relative standard deviation of 10 measurements of resistant starch content in the sample was less than 10%,indicating good precision of the method. The error between the measured value and the labeled value of the quality control sample with known resistant starch content was 2.62%,indicating high accuracy of the method. In conclusion,the method established in this ar‐ticle is suiTablefor the determination of resistant starch content in food.
