中文摘要: 为研究富含羟基酪醇和玉米黄质色素的橄榄油对碘酸钠诱导视力障碍小鼠的影响,对小鼠腹腔注射浓度为5%的碘酸钠,构建视网膜损伤的视力障碍小鼠动物模型,将建模成功的小鼠分为模型组、橄榄油(olive oil,OL)组、富含羟基酪醇橄榄油(olive oil rich in hydroxyl tyrosol,OHT)组、富含玉米黄质色素橄榄油(olive oil rich in zeaxanthin pigment,OZT)组、安多明药物(calcium dobesilate capsules,CDC)组,以生理盐水为对照(CK)组。连续灌胃28 d,分析视力障碍小鼠视网膜的变化和OZT、OHT 对视网膜中过氧化氢酶(catalase,CAT)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH‐Px)和超氧化物歧化酶(superoxide dismutase,SOD)的活性以及丙二醛(malondialdehyde,MDA)、白细胞介素‐12(interleukin‐12,IL‐12)、白细胞介素‐18(interleukin‐18,IL‐18)、肿瘤坏死因子‐a(tumor necrosis factor‐α,TNF‐a)和转化生长因子‐b(transforming growth factor‐β,TGF‐b)含量的影响。结果表明,富含羟基酪醇橄榄油和富含玉米黄质色素橄榄油均能有效增加色素上皮衍生因子(serpin peptidase inhibitor clade F member1,SERPINF1)和色素上皮衍生因子受体(recombinant patatin like phospholipase domain containing protein 2,PNPLA2)阳性表达量,升高小鼠视网膜中CAT、GSH‐Px 和SOD 活性,降低MDA、IL‐12、IL‐18、TNF‐a 和TGF‐b 含量,恢复小鼠视网膜厚度和细胞数量,对小鼠视网膜损伤有很好的保护作用,且富含玉米黄质色素橄榄油效果优于富含羟基酪醇橄榄油。综上,OZT 和OHT对视力障碍小鼠视网膜损伤具有很好的保护和辅助治疗效果。
Abstract:To study the effects of olive oil rich in hydroxytyrosol and zeaxanthin pigment on visual impairment induced by sodium iodate in mice,a mice model of visual impairment featured by retinal damage was estab‐lished by intraperitoneal injection of sodium iodate at a concentration of 5%. Successfully modeled mice were divided into the following groups:model group,olive oil(OL)group,olive oil rich in hydroxy tyrosol(OHT)group,olive oil rich in zeaxanthin pigment(OZT)group,calcium dobesilate capsules(CDC)group,with a physiological saline group as the control group(CK). After 28 consecutive days of gavage administration,changes in the retina of mice with visual impairment were analyzed,and the effects of OZT and OHT on the changes of the activities of catalase(CAT),glutathione peroxidase(GSH‐Px)and superoxide dismutase(SOD),as well as the content of malondialdehyde(MDA),interleukin‐12(IL‐12),interleukin‐18(IL‐18),tu‐mor necrosis factor‐α(TNF‐α)and transforming growth factor‐β(TGF‐β)in the retina were evaluated.The re‐sults showed that both olive oil rich in hydroxytyrosol and olive oil rich in zeaxanthin pigment effectively in‐creased the positive expression levels of serpin peptidase inhibitor clade F member 1(SERPINF1)and recom‐binant patatin like phospholipase domain containing protein 2(PNPLA2),increased the activities of CAT,GSH‐Px,and SOD in the retina of mice,and reduced the content of MDA,IL‐12,IL‐18,TNF‐α,and TGF‐β.Additionally,they recovered the retinal thickness and cell count in mice,providing significant protective ef‐fects against retinal damage.Notably,olive oil rich in zeaxanthin pigment exhibited superior efficacy compared to olive oil rich in hydroxytyrosol. In conclusion,OZT and OHT have good protective and auxiliary therapeutic effects on mice with visual impairment featured by retinal damage.
