中文摘要: 该文针对生物催化剂——黑曲霉 C2J6 全细胞,比较培养基组成对全细胞催化合成能力的影响,通过单因素试验和响应面优化试验,确定非水相中全细胞催化合成乳酸乙酯的最佳条件。结果表明,菌株在液体菌种培养基中,28 ℃、150 r/min 下培养24 h,全细胞的催化活性最高;合成乳酸乙酯的最佳条件为反应介质叔丁醇5 mL、底物(乳酸和无水乙醇)物质的量比1∶5、乳酸浓度0.3 mol/L(以反应介质体积为基准)、全细胞浓度35 g/mol(以乳酸物质的量为基准),在40 ℃、150 r/min 下反应24 h,乳酸乙酯产率最高,达29.86%。全细胞催化下的乳酸乙酯合成反应符合准一级动力学模型,全细胞重复使用3 次后的催化效率逐渐下降,酯化率仅剩2.90%。
Abstract:In this paper, the effects of medium composition on the catalysis ability of whole cells of Aspergillus niger C2J6 biocatalyst were compared, and the optimal conditions for the synthesis of ethyl lactate by whole cell catalysis in a non-aqueous phase were determined by single factor test and response surface optimization test. The results showed that the whole cell had the highest catalytic activity when cultured at 28 ℃ and 150 r/min for 24 h in a liquid culture medium. The optimal conditions for synthesis of ethyl lactate were as follows: reaction medium tert-butanol of 5 mL, substrate molar ratio (lactic acid and anhydrous ethanol) of 1∶5, lactic acid concentration of 0.3 mol/L (based on the volume of the reaction medium), whole cell concentration of 35 g/mol (based on the amount of lactic acid substance), and reaction at 40 ℃ and 150 r/min for 24 h. The yield of ethyl lactate was the highest (29.86%). The synthesis of ethyl lactate by whole cell catalysis was consistent with the quasi-first-order kinetic model, and the catalytic efficiency decreased gradually after the whole cell was reused for three times, and the esterification rate was only 2.90%.
