中文摘要: 为研究益生核桃钙对骨质疏松大鼠骨密度的改善作用及其对肝脏代谢的影响,将40 只大鼠随机分为模型(MC)组、阳性对照(CA)组、益生核桃钙高(WCH)、中(WCM)、低剂量(WCL)组,采用营养干预造模法造模,3 个月后,进行骨相关参数和肝脏代谢组学检测分析。结果表明,与MC 组相比,CA 及WCH 组血清钙含量、骨强度、骨钙含量及骨密度显著增高(P<0.05),且WCH 组的升高幅度及显著水平均优于CA 组。与MC 组相比,WCH 组和CA 组在肝脏代谢水平上涉及的差异代谢物分别为145 个和144 个,其中38 个相同代谢物的上调和下调趋势相同。WCH 组与CA 组相比有107 个差异显著的代谢物,提示益生核桃钙和碳酸钙对肝脏代谢的影响不完全相同。MC 组、WCH组和CA 组间的大部分差异代谢物富集在嘌呤、能量和神经相关代谢通路以及FoxO 信号通路,甲状旁腺激素的合成、分泌和作用,成瘾代谢和蛋白质消化吸收等。提示益生核桃钙含有有机钙、核桃蛋白和益生元,这些成分能够共同作用于相关代谢途径,调节机体肝脏的代谢功能,有助于促进钙吸收、提高股骨的骨强度、达到改善骨质疏松的作用。
Abstract:The effects of prebiotic walnut calcium on the bone mineral density and liver metabolism in the rat model of osteoporosis were investigated. Forty rats were randomly grouped as follows:model (MC),positive control (CA),and high-,medium-,and low-dose prebiotic walnut calcium(WCH,WCM,and WCL,respectively). The nutrition intervention method was employed to establish the rat model of osteoporosis. After three months,bone-related parameters and liver metabolomics were analyzed. The results showed that compared with the MC group,the CA and WCH groups showed increases in serum calcium content,bone strength,bone calcium content,and bone mineral density (P<0.05),and the increases in the WCH group were higher than those in the CA group. Compared with the MC group,the WCH and CA groups involved 145 and 144 differential metabolites,respectively,at the liver metabolism level,among which 38 common metabolites showed the same upregulation and down-regulation trends. There were 107 differential metabolites between the WCH group and the CA group,which suggested that prebiotic walnut calcium and calcium carbonate had different effects on liver metabolism. Most of the differential metabolites between MC,WCH,and CA groups were concentrated in purine,energy,and neurorelated metabolic pathways,FoxO signaling pathway,synthesis,secretion,and action of parathyroid hormone,addiction metabolism,and protein digestion and absorption. The product contained organic calcium,walnut protein,and prebiotics,which acted together on relevant metabolic pathways to regulate liver metabolism. It helped to promote calcium absorption,increase femur bone strength,and ameliorate osteoporosis.
