中文摘要: 该研究旨在探讨茯苓薏苡仁水提物对高脂饮食诱导的肥胖小鼠的影响,采用高脂饲料喂养小鼠建立肥胖模型,连续给药12 周,监测每周体质量、饮食饮水。实验结束后,检测小鼠生理生化指标,采用蛋白免疫实验检测脂肪组织脂解和褐变的相关蛋白表达情况。结果表明,与高脂组相比,茯苓薏苡仁水提物明显抑制小鼠体质量和脂肪质量的增加,降低血脂水平,改善糖脂代谢紊乱,增加腺苷酸活化蛋白激酶(AMP-activated protein kinase,AMPK)、解偶联蛋白1(uncoupling protein 1,UCP-1)、过氧化物酶体增殖物活化受体γ 共激活因子-1α(peroxisome proliferatoractivated receptor γ coactivator-1α,PGC-1α)、甘油三酯脂肪酶(adipose triglyceride lipase,ATGL)、激素敏感性脂肪酶(hormone-sensitive lipase,HSL)的相对表达量。综上,茯苓薏苡仁水提物能够减轻小鼠体质量、减少脂肪堆积、改善糖脂代谢紊乱,其作用机制与促进脂解和白色脂肪褐变相关。

Abstract:This study investigated the effect of an aqueous extract of Poria cocos and Coix seed on obese mice induced by a high-fat diet (HFD). Mice were fed an HFD to establish an obesity model. They were administered drugs for 12 weeks,and their body mass,diet,and drinking water were monitored every week. After the experiment,the physiological and biochemical indexes of mice were detected. Western blotting assay was used to detect the expression of proteins related to lipolysis and browning in adipose tissue. The results showed that compared with the HFD group,the aqueous extract of Poria cocos and Coix seed significantly inhibited the increase in the body weight and fat mass in mice,reduced blood lipid levels,improved the disorders of glucose and lipid metabolism,and increased the relative expression levels of AMP-activated protein kinase(AMPK),uncoupling protein 1 (UCP-1),peroxisome proliferator-activated receptor γ coactivator-1α (PGC-1α),adipose triglyceride lipase (ATGL),and hormone-sensitive lipase (HSL). In conclusion,the aqueous extract of Poria cocos and Coix seed could reduce body weight,reduce fat accumulation,and improve glucose and lipid metabolism disorders in mice,and its mechanism of action was related to promoting lipolysis and white fat browning.
