中文摘要: 为提高小麦资源的利用率,该试验采用双酶分步酶解法处理小麦面筋蛋白,研究4 种蛋白酶对小麦面筋蛋白蛋白质得率的影响,筛选出蛋白质得率较高的蛋白酶进行单因素试验和正交试验,并进一步优化双酶分步酶解顺序和时间。结果表明,最优工艺条件:首先采用中性蛋白酶酶解,底物浓度10%,pH 值为7.0,酶添加量为8 000 U/g,酶解温度55 ℃,酶解时间1.5 h;然后在此基础上进行碱性蛋白酶酶解,调节pH 值至8.0,酶添加量为8 000 U/g,酶解温度60 ℃,酶解时间1.5 h。基于此工艺条件下得到酶解液的蛋白质得率在90%以上。
Abstract:To enhance the utilization of wheat resources, a two-step enzymatic hydrolysis method was employed to process wheat gluten. The effects of four proteases on the protein yield from wheat gluten were investigated. Protease, which resulted in higher protein yields, was selected for single-factor and orthogonal tests,which was conducted to optimze the two-step enzymatic hydrolysis sequence and duration. The results indicated that the optimal conditions of the first step was neutral protease hydrolysis at pH7.0, enzyme addition of 8 000 U/g, substrate concentration of 10%, hydrolysis temperature of 55 ℃, and hydrolysis time of 1.5 h. The second step was alkaline protease hydrolysis at pH8.0, enzyme addition of 8 000 U/g, hydrolysis temperature of 60 ℃, and hydrolysis time of 1.5 h. Under the optimized conditions, the protein yield exceeded 90%.
