中文摘要: 以β-葡聚糖得率得率为主要评价指标,通过单因素和响应面试验优化蛹虫草β-葡聚糖超声辅助水提工艺,采用红外扫描分析提取物的化学键及官能团信息,测定其中各组分含量及其体外抗氧化活性。结果表明,蛹虫草β-葡聚糖最优提取工艺为超声时间1 h、料液比1∶31(g/mL)、水浴温度90 ℃。最优条件下提取得率为(1.94±0.28)%,与预测值1.95% 一致。经过纯化试验得出α-葡聚糖去除最优条件为α-淀粉酶添加量2%、孵育时间60 min、温度50 ℃,经过纯化处理后β-葡聚糖纯度由19.82% 提高至33.93%;红外扫描结果表明,蛹虫草β-葡聚糖具有典型的多糖结构,并含有硫酸根基团。蛹虫草β-葡聚糖对DPPH 自由基、ABTS+自由基和羟基自由基的清除能力与浓度呈正相关(2.00~10.00 mg/mL),IC50 值分别为1.17、2.17、4.35 mg/mL,清除率在浓度为8.34、9.88、9.97 mg/mL 时,可达100%。综上,优化后的蛹虫草β-葡聚糖超声辅助水提工艺稳定可行,提取获得的β-葡聚糖具有较好的抗氧化活性。
Abstract:The ultrasound-assisted extraction process of β-glucan from Cordyceps militaris was optimized by single factor experiment and response surface methodology with β-glucan extraction yield as the main evaluation index.The chemical bonds and functional groups of the extracts were analyzed by infrared scanning,and the content of each component and its antioxidant activity in vitro were determined.The results showed that the optimal extraction process of β-glucan from C.militaris was ultrasonication time of 1 h,material-liquid ratio of 1∶31(g/mL),and temperature of 90°C.The extraction rate under the optimal conditions was(1.94±0.28)%,which was consistent with the predicted value of 1.95%.The purification test revealed that the optimal conditions for α-glucan removal were α-amylase addition of 2%,incubation time of 60 min,and temperature of 50 ℃.The purity of β-glucan increased from 19.82% to 33.93% after the purification treatment.The result of infrared scanning showed that C.militaris β-glucan had a typical polysaccharide structure with sulfate moieties.The scavenging abilities of C.militaris β-glucan on DPPH radicals,ABTS+ radicals,and hydroxyl radicals were positively correlated with the concentration(2.00-10.00 mg/mL),with IC50 values of 1.17,2.17 mg/mL and 4.35 mg/mL respectively.The scavenging rate was 100% at the concentration of 8.34,9.88 and 9.97 mg/mL.In conclusion,the optimized ultrasound-assisted extraction process of β-glucan from C.militaris was stable and feasible,and the antioxidant activity of the β-glucan extracted was good.
