中文摘要: 该文分别以水和60%乙醇为提取溶剂,在55、75 ℃和95 ℃下对沙棘叶茶整叶和沙棘叶茶粉进行提取,比较提取物中总黄酮、可溶性总多酚、异鼠李素含量及抗氧化能力和α-糖苷酶抑制能力。结果表明,提取溶剂为60%乙醇、提取温度为95 ℃时,沙棘叶茶粉提取物中的活性成分含量最高,总黄酮、可溶性总多酚、异鼠李素含量分别为(82.59±1.80)、(117.02±1.93)、(1.592±0.074)mg/g,清除DPPH 自由基、羟基自由基和亚硝酸根的能力最强,其EC50值分别为(0.141 9±0.000 6)、(0.357 1±0.004 8)、(0.815 6±0.001 3)mg/g,还原力最高为(118.65±2.40)mg/g,α-糖苷酶抑制能力最强,IC50值为(0.393 0±0.010 1)mg/g。通过主成分分析,该提取物的样品评分最高。
Abstract:The whole leaves and powder of sea buckthorn leaf tea were extracted with water and 60% ethanol at 55,75 ℃,and 95 ℃,respectively.The total flavonoid content,total soluble polyphenol content,isorhamnetin content,antioxidant capacity,and α-glucosidase inhibitory activity were compared between different extracts.The results showed that extract of sea buckthorn leaf tea powder obtained by extraction with 60% ethanol at 95 ℃had the highest content of total flavonoids,total soluble polyphenols,and isorhamnetin,which was(82.59±1.80),(117.02±1.93)mg/g,and(1.592±0.074)mg/g,respectively.This extract showed the strongest scavenging activities against DPPH free radicals,hydroxyl free radicals,and nitrite,with the EC50 values of(0.141 9±0.000 6),(0.357 1±0.004 8)mg/g,and(0.815 6±0.001 3)mg/g,respectively.In addition,it demonstrated the strongest reducing power [(118.65±2.40)mg/g] and the strongest inhibitory activity against α-glycosidase with the IC50 of(0.393 0±0.010 1)mg/g.The results of principal component analysis showed that the 60% ethanol extract obtained at 95 ℃had the highest comprehensive score.
