中文摘要: 采用形态学观察及内部转录间隔区(internal transcribed spacer,ITS)序列分析的方法,对1株寄生于桑树的野生粗毛纤孔菌(SK-4)进行鉴定,使用单因子试验方法确定菌丝最适培养条件,并研究子实体不同提取物中的黄酮、多酚、多糖的含量及其抗氧化活性。结果表明:野生菌株SK-4 为粗毛纤孔菌Inonotus hispidus,其菌丝体生长最适氮源为酵母粉,最适碳源为蔗糖,最适pH 值为5.0,最适生长温度为25~28 ℃。SK-4 菌株子实体的水提物及醇提物中多糖、黄酮及多酚的含量及抗氧化活性差异明显,其中70%乙醇提取物抗氧化活性明显优于水提物,子实体多糖主要存在水提物中,多糖的含量与1,1-二苯基-2-苦基肼(1,1-diphenyl-2-picrylhydrazyl,DPPH)自由基清除能力呈中等正相关(R2=0.557),与总抗氧化能力、羟自由基清除率呈负相关,70%乙醇溶液有利于黄酮、多酚的提取,二者的含量与总抗氧化能力、羟自由基清除率呈极显著正相关(R2>0.95),与DPPH 自由基清除能力呈负相关。
Abstract:A wild strain(SK-4)parasitizing mulberry in Liaoning was identified based on the morphological characteristics and internal transcribed spacer(ITS)sequence.The optimal culture conditions were determined by single factor experiments,and different extracts of fruiting bodies were measured in terms of the content of flavonoids,polyphenols,and polysaccharide and antioxidant activity.The strain SK-4 was identified as Inonotus hispidus.The optimal conditions for the mycelial growth were sucrose as the carbon source,yeast powder as the nitrogen source,pH5.0,and 25-28 ℃.The content of polysaccharides,flavonoids,and polyphenols and the antioxidant activity were significantly different between the ethanol extract and the water extract.The 70% ethanol extract showed significantly higher antioxidant activity than the water extract.The polysaccharide of fruiting bodies mainly existed in the water extract.The content of polysaccharide had a moderately positive correlation with the 1,1-diphenyl-2-picrylhydrazyl(DPPH)free radical scavenging activity(R2=0.557)and negative correlations with total antioxidant capacity and hydroxyl free radical scavenging activity.The 70% ethanol solution was suitable for the extraction of flavonoids and polyphenols,the content of which had positive correlations with the total antioxidant capacity and hydroxyl radical scavenging rate(R2>0.95)and a negative correlation with the DPPH free radical scavenging activity.
