中文摘要: 为探究仙草提取物对大肠杆菌的抑制作用,测定其对大肠杆菌的抑菌圈大小、最小抑菌浓度(minimal inhibitory concentration,MIC)和最小杀菌浓度(minimum bactericidal concentration,MBC),通过液相色谱测定其抑菌成分的含量和对应的MIC 和MBC;通过碱性磷酸酶试验测定其对细胞壁通透性的影响,通过测定电导率和OD260 nm 变化,分析其对细胞膜完整性和通透性的影响。结果表明:仙草提取物的抑菌圈为(21.69±2.15)mm,最小抑菌浓度为0.17 g/mL(生药浓度),最小杀菌浓度为0.33 g/mL。提取物中芦丁、异槲皮苷、紫云英苷、迷迭香酸和咖啡酸的含量分别为6.63、0.47、0.34、0.32、0.01 mg/g,迷迭香酸、咖啡酸、芦丁、异槲皮苷、紫云英苷、齐墩果酸、熊果酸的MIC 分别为4.78、5.10、6.05、1.61、0.84、0.34、0.33 mg/mL,其对应的MBC 分别为9.55、10.20、12.10、3.21、1.68、0.68、0.65 mg/mL。仙草提取物能通过增加细胞壁、细胞膜的通透性及破坏其细胞膜的完整性,有效抑制大肠杆菌的生长。

Abstract:The inhibitory effect of the extract from Hsian-tsao(HST)on Escherichia coli(E.coli)was explored.The inhibition zone diameter,minimal inhibitory concentration(MIC),and minimum bactericidal concentration(MBC)of the extract from HST on the E.coli were measured.The content of antibacterial components and corresponding MIC and MBC were determined by liquid chromatography.The effect on cell wall permeability was determined by alkaline phosphatase assay,and that on cell membrane integrity and permeability was analyzed by measuring the conductivity and OD260 nm changes.The results showed that the inhibition zone diameter of the extract from HST was(21.69±2.15)mm.The MIC was 0.17 g/mL(drug concentration),and MBC was 0.33 g/mL.The contents of rutin,isoquercitrin,astragalin,rosmarinic acid,and caffeic acid were 6.63,0.47,0.34,0.32 mg/g and 0.01 mg/g,respectively in the extract.The MIC of rosmarinic acid,caffeic acid,rutin,isoquercitrin,astragalin,ursolic acid,and oleanolic acid was 4.78,5.10,6.05,1.61,0.84,0.34 mg/mL and 0.33 mg/mL,respectively.The corresponding MBC was 9.55,10.20,12.10,3.21,1.68,0.68 mg/mL and 0.65 mg/mL.The extract from SHT could effectively inhibit the growth of E.coli by increasing the permeability of the cell wall and cell membrane and disrupting the integrity of the cell membrane.
