中文摘要: 为探讨山药硒多糖(Chinese yam selenium polysaccharide,SeY)对镉(Cd)暴露小鼠Cd 蓄积和氧化应激的缓解作用,选取100 只成年C57BL/6 雄性小鼠,适应性饲养1 周后,随机分成5 组[对照组(CK 组),氯化镉处理组(Cd组),山药硒多糖低(L-SeY)、中(M-SeY)、高剂量组(H-SeY)],6 周后收集组织器官和血液进行测定。结果显示,Cd在小鼠各器官的蓄积分别为肾脏(47.55 μg/g)>肝脏(23.15 μg/g)>心脏(13.26 μg/g)>小肠(9.31 μg/g)>大脑(0.07 μg/g),血液中Cd 含量为5.17 μg/L。此外,Cd 组小鼠肝脏指数显著升高,且Cd 暴露显著提高了小鼠血清中谷草转氨酶(aspartate transaminase,AST)、谷丙转氨酶(alanine aminotransferase,ALT)、乳酸脱氢酶(lactate dehydrogenase,LDH)和碱性磷酸酶(alkaline phosphatase medicine,ALP)活性,小鼠肝脏中超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)、过氧化氢酶(catalase,CAT)和谷胱甘肽还原酶(glutathione reductase,GR)活性显著降低,而丙二醛(malondialdehyde,MDA)水平显著升高(P<0.05)。而山药硒多糖显著降低了Cd 暴露小鼠血液、肾脏、肝脏、小肠和心脏中的Cd 含量(P<0.05)。此外,山药硒多糖显著降低了Cd 暴露小鼠血清中AST、ALT、LDH 和ALP 活性(P<0.05),显著升高了小鼠肝脏中SOD、GSH-Px、CAT 和GR 活性,并降低了MDA 水平(P<0.05),且呈剂量依赖性。综上,山药硒多糖可降低Cd 在小鼠各组织器官中蓄积并缓解Cd 暴露引起的小鼠氧化应激。
Abstract:To investigate the alleviating effects of Chinese yam selenium polysaccharide(SeY)on cadmium(Cd)accumulation and oxidative stress in Cd-exposed mice,100 C57BL/6 adult male mice were selected.After 1 week of adaptive feeding,they were randomly divided into 5 groups,namely the control group(CK group),the cadmium chloride treatment group(Cd group),the Chinese yam selenium polysaccharide with low(L-SeY),medium(M-SeY),and high-dose groups(H-SeY).After 6 experimental weeks,their tissues,organs,and blood were collected for experimental determination.The results showed that the Cd cumulant in various organs of mice was kidney(47.55 μg/g)>liver(23.15 μg/g)>heart(13.26 μg/g)>small intestine(9.31 μg/g)>brain(0.07 μg/g),and the Cd content in blood was 5.17 μg/L.In addition,the liver index of mice in the Cd group was significantly increased.In addition,Cd exposure significantly increased the levels of aspartate transaminase(AST),alanine aminotransferase(ALT),lactate dehydrogenase(LDH),and alkaline phosphatase medicine(ALP)in mice serum,significantly decreased the levels of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),catalase(CAT)and glutathione reductase(GR)in the mice liver,and significantly increased the malondialdehyde(MDA)level(P<0.05).However,Chinese yam selenium polysaccharide significantly decreased the Cd content in the blood,kidney,liver,small intestine,and heart of Cd-exposed mice(P<0.05),significantly decreased the levels of AST,ALT,LDH,and ALP in the serum of Cd-exposed mice(P<0.05).It also significantly increased the levels of SOD,GSH-Px,CAT,and GR in the mice liver,and decreased MDA levels(P<0.05)in a dose-dependent manner.To sum up,the above results showed that Chinese yam selenium polysaccharide can reduce the Cd accumulation in various tissues and organs of mice and alleviate the oxidative stress in mice caused by Cd exposure.
