茶条槭叶没食子酸提取工艺优化及抗氧化活性

中文摘要: 为探索茶条槭叶没食子酸的提取技术,以确立其最佳提取工艺,以单宁酶耦联超声-微波协同提取为方法,60%乙醇溶液为提取溶剂,在提取时间、单宁酶添加量、微波功率、提取温度、料液比以及pH 值6 个单因素试验的基础上,以没食子酸得率为评价指标,通过响应面法对提取工艺进行优化,采用离子交换树脂重结晶的方法进一步纯化没食子酸。以DPPH 和ABTS+自由基清除率为指标,评价没食子酸和抗坏血酸(vitamin,VC)的体外抗氧化活性。结果表明,茶条槭叶没食子酸的最佳提取工艺参数为提取时间40 min、单宁酶添加量4.3%、料液比1∶60(g/mL)、微波功率520 W、提取温度60 ℃、pH7。在此条件下没食子酸得率为(26.25±0.41)%,与理论推测值26.64%接近,经纯化后没食子酸的纯度达到90%以上。在DPPH 和ABTS+自由基清除试验中,茶条槭叶没食子酸的清除率均比VC 的清除率高。研究表明单宁酶耦联超声-微波协同提取法能够提高茶条槭叶没食子酸得率,茶条槭叶没食子酸具有较强的抗氧化能力。
Abstract:This study aimed to explore the extraction technique of gallic acid from Acer ginnala leaves to determine its optimal extraction process.The extraction process was optimized by response surface methodology by the tannase-coupled ultrasound-microwave synergistic extraction method with 60% ethanol solution as the extraction solvent and the yield of gallic acid as the evaluation index based on the six single factor tests,including extraction time,tannase addition,microwave power,extraction temperature,solid-to-liquid ratio,and pH value.Gallic acid was further purified using the ion exchange resin recrystallization method.The in vitro antioxidant activities of gallic acid and ascorbic acid(VC)were evaluated with DPPH and ABTS+ radical scavenging rates as indicators.The results showed that the optimal extraction process parameters for gallic acid were as follows: extraction time of 40 min,tannase addition of 4.3%,solid-to-liquid ratio of 1∶60(g/mL),microwave power of 520 W,extraction temperature of 60 ℃,and pH value of 7.Under these conditions,the yield of gallic acid was(26.25±0.41)%,which was close to the theoretically predicted value of 26.64%.After purification,the purity of gallic acid reached over 90%.In DPPH and ABTS+ free radical scavenging experiments,the scavenging rate of gallic acid in Acer ginnala leaves was higher than that of VC.The results have shown that the tannase-coupled ultrasound -microwave synergistic extraction method can improve the yield of gallic acid in Acer ginnala leaves which has strong antioxidant capacity.
