中文摘要: 为探究低、中、高剂量金丝桃苷对酒精饮食诱发的小鼠酒精性肝病的影响,该文建立短期酒精性脂肪肝炎模型(Gao-Binge 模型),测定血清转氨酶、甘油三酯(triglyceride,TG)含量,以及肝组织中超氧化物歧化酶(superoxide dismutase,SOD)活性、丙二醛(malondialdehyde,MDA)含量,并对肝脏进行苏木精-伊红染色(hematoxylin and eosin staining,H&E)和油红染色观察肝脏组织病理学变化,免疫组化染色检测肝脏中性粒细胞浸润情况,最后利用实时定量聚合酶链式反应(real-time quantitative polymerase chain reaction,RT-PCR)技术分析肝脏炎性细胞因子和趋化因子的表达。结果显示,酒精液体饲料喂养后,模型组小鼠出现脂肪性肝炎,而金丝桃苷能够降低血清转氨酶和TG 含量,减少肝组织油红染色区域,降低肝脏MDA 含量,提升SOD 活性。此外,金丝桃苷抑制酒精饮食诱发的肝脏白细胞介素6(interleukin 6,IL-6)和肿瘤坏死因子(tumor necrosis factor,TNF-α)表达,减少肝组织中性粒细胞浸润及其趋化因子表达,表明金丝桃苷可以通过降低肝脏脂质过氧化水平,抑制炎症反应来改善小鼠酒精性脂肪肝炎。
Abstract:A short-term alcoholic steatohepatitis model(Gao-Binge model)was established to explore the effects of hyperin with low,moderate,or high dosages on alcoholic liver diseases induced by the alcoholic diet of mice.Levels of serum transaminase,triglyceride(TG),superoxide dismutase(SOD),and malondialdehyde(MDA)in liver tissue were measured.Liver histopathological changes were observed by hematoxylin and eosin staining(H&E staining)and oil red staining.Liver neutrophil infiltration was detected by immune-histochemical staining.Finally,the expression of inflammatory cytokines and chemokines in liver was analyzed by real-time quantitative polymerase chain reaction(RT-PCR).The results showed that after feeding with an alcohol liquid diet,the mice in the model group developed steatohepatitis,while hyperin could significantly reduce the levels of serum transaminase and TG and the area of oil red staining,decrease the level of MDA in liver,and increase the activity of SOD enzyme.In addition,hyperin inhibited the expression of interleukin 6(IL-6)and tumor necrosis factor(TNF-α)in liver induced by an alcoholic diet and reduced neutrophil infiltration and chemokine expression in liver tissue.This suggested that hyperin could improve alcoholic steatohepatitis in mice by reducing the level of lipid peroxidation in the liver and inhibiting inflammation.
