中文摘要: 该文对快速检测食品中环丙沙星残留的生物条形码检测技术进行研究。首先将环丙沙星多克隆抗体与条形码DNA 链共同修饰在纳米金颗粒上制备纳米金复合探针、环丙沙星单克隆抗体与磁性颗粒结合制备磁性探针,然后对其采用可见光光谱法、透射电镜法进行鉴定;其次将上述探针与环丙沙星标准品进行杂交反应,在磁场的作用下,去除未结合的纳米金复合探针,然后收集标记成功的纳米金复合探针释放出的DNA 链,采用微孔板银染的生物条形码技术进行检测,得到环丙沙星残留量。检测环丙沙星的最低检出限为2.13 ng/mL。该方法与常规免疫检测方法相比灵敏度增强,具有实用性,为未来检测小分子物质残留量提供了新思路,也为制备环丙沙星生物条形码检测试剂盒奠定了基础。
Abstract:This study investigates the biobarcode detection technology for rapid detection of enrofloxacin residues in food. Firstly,enrofloxacin polyclonal antibodies and barcode DNA strands are immobilized on gold nanoparticles to prepare nanocomposite probes,while enrofloxacin monoclonal antibodies are conjugated to magnetic particles to prepare magnetic probes. The probes are characterized using visible light spectroscopy and transmission electron microscopy. Then,the probes are hybridized with enrofloxacin standard samples,and unbound nanocomposite probes are removed using a magnetic field. The released DNA strands from the successfully labeled nanocomposite probes are collected,and the residual enrofloxacin is detected using the biological barcode technique with silver staining on a microtiter plate. The method achieves a minimum detection limit of 2.13 ng/mL for enrofloxacin. Compared to conventional immunoassay methods,this approach enhances sensitivity and demonstrates practicality,providing a new strategy for the detection of small molecule residues and laying the foundation for the development of enrofloxacin biobarcode detection kits in the future.
