中文摘要: 为比较乙醇/硫酸铵和乙醇/磷酸二氢钠两种双水相体系对蓝莓皮渣粗提液中的花青素的纯化效果,通过单因素试验和正交试验,得到最佳的工艺条件:乙醇/硫酸铵体系中硫酸铵质量分数22.5%、乙醇质量分数24.0%、pH4.5、色素添加量15%,选择性系数β 为27.10;乙醇/磷酸二氢钠体系中磷酸二氢钠质量分数25%、乙醇质量分数29%、pH2.5、色素添加量14%,选择性系数β 为140.05,乙醇/磷酸二氢钠体系的选择性系数是乙醇/硫酸铵体系的5 倍。采用乙醇/磷酸二氢钠体系纯化的色素保留率高,且磷酸二氢钠具有溶解度低、易于回收的特点,所以该体系具有更好的工业化应用前景。
Abstract:Ethanol/ammonium sulfate and ethanol/sodium dihydrogen phosphate aqueous two-phase systems were compared in the purification of anthocyanins from blueberry peel residues.The single factor and orthogonal tests were conducted to optimize thepurification conditions.The ethanol/ammonium sulfate system was optimized as ammonium sulfate of 22.5%,ethanol of 24.0%,pH4.5,and pigment addition of 15%,under which the selectivity coefficient β was 27.10.The ethanol/sodium dihydrogen phosphate system was optimized as sodium dihydrogen phosphate of 25%,ethanol of 29%,pH2.5,pigment addition of 14%,under which the selectivity coefficient β was 140.05.The selectivity coefficient of the ethanol/sodium dihydrogen phosphate system was 5 times that of the ethanol/ammonium sulfate system.The anthocyanins purified by the ethanol/sodium dihydrogen phosphate system had a high retention rate.Moreover,sodium dihydrogen phosphate in the system had low solubility and was easy to be recovered.Therefore,this system demonstrated a promosing prospect of industrial application.
