一株低温蛋白酶产生菌的分离及该酶的鉴定与酶学性质表征

中文摘要: 从低温环境中分离并鉴定出1 株产低温蛋白酶的乙酰微小杆菌YD37(Exiguobacterium acetylicum YD37)。对该低温蛋白酶基因序列分析发现,其基因全长1 110 bp,编码370 个氨基酸残基,分子量为41.5 kDa,属于氨肽酶,富含短侧链和带负电荷的氨基酸残基,二级结构含有14 个α-螺旋和19 个β-折叠,三级结构表面为带负电荷氨基酸构成的亲水表面。通过对E.acetylicum YD37 发酵产酶的工艺进行优化,酶活提高8.26 倍,达到273.5 U/mL。经硫酸铵沉淀和离子交换层析得到该低温蛋白酶的粗酶。粗酶的最适温度为35 ℃,但20 ℃仍能保持43.6%的酶活。酶活受到Mn2+和Co2+的促进和乙二胺四乙酸的完全抑制,表明该酶是一种金属氨肽酶。该酶的动力学常数Km 和Vmax 分别为1.38 mg/mL 和4.41 mg/(mL·min)。
Abstract:Exiguobacterium acetylicum YD37 producing cold-adaptive protease was isolated from a lowtemperature environment and identified.The cold-adaptive protease gene had a full length of 1 110 bp,which encoded a 41.5 kDa protein composed of 370 amino acid residues.This protease was an aminopeptidase with rich short side chains and negatively charged residues.The secondary structure of the protease contained 14 αhelixes and 19 β-sheets,and the three-dimensional structure had a hydrophilic surface with rich negatively charged residues.The enzyme activity was increased by 8.26 folds to 273.5 U/mL after optimization of the fermentation process of E.acetylicum YD37.The crude protease was purified by ammonium sulfate precipitation and ion exchange chromatography.The protease showed the highest activity at 35 ℃,while it still maintained a relative activity of 43.6% at 20 ℃.The enzyme was indicated as a metalloprotease since its activity was stimulated by Mn2+and Co2+but inhibited completely by enthylene diamine tetraacetic acid.Its Km and Vmax was calculated to be 1.38 mg/mL and 4.41 mg/(mL·min),respectively.
