产环糊精葡萄糖基转移酶苦橙内生细菌的分离鉴定

中文摘要: 为寻找产环糊精葡萄糖基转移酶(cyclodextrin glycosyltransferase,CGTase)的微生物新菌株,采用酚酞-甲基橙筛选平板、淀粉-碘筛选平板从苦橙内生细菌中分离获得一株可产环糊精葡萄糖基转移酶的新菌株,命名为KCEB04。形态学特征、生理生化反应、16S rDNA 序列同源性分析结果表明,菌株KCEB04 归属于产酸克雷伯氏菌(Klebsiella oxytoca)。酶活测定结果表明,该菌株所产淀粉水解酶酶活为5 884.35 U/mL、α-CGTase 酶活为1.65 U/mL、β-CGTase 酶活为0.44 U/mL、γ-CGTase 酶活为1.05 U/mL。
Abstract:To search for new microbial strains producing cyclodextrin glucosyltransferase(CGTase),a strain capable of producing CGTase was isolated from endophytic bacteria of bitter orange using phenolphthaleinmethyl orange screening plates and starch-iodine screening plates,and named KCEB04.Morphological characteristics,physiological and biochemical reactions,and 16S rDNA sequence homology analysis results showed that strain KCEB04 belonged to Klebsiella oxytoca.Enzyme activity assay results showed that the strain produced 5 884.35 U/mL amylase,1.65 U/mL α-CGTase,0.44 U/mL β-CGTase,and 1.05 U/mL γ-CGTase.
