中文摘要: 该文研究常见类胡萝卜素对HepG2 细胞紫外氧化损伤的影响。通过HepG2 细胞毒性试验、紫外线A(ultraviolet A,UVA)辐照试验、还原型谷胱甘肽(glutathione,GSH)含量测定和总抗氧化能力(total antioxidant capacity,TAOC)分析等,研究常见类胡萝卜素(法夫酵母类胡萝卜素提取物、虾青素、β-胡萝卜素和海胆酮)对HepG2 细胞氧化损伤的保护作用。结果表明,虾青素、β-胡萝卜素和海胆酮均能提高UVA 辐照损伤的HepG2 细胞的存活率、GSH 含量和T-AOC,其中虾青素对HepG2 细胞的活性氧(reactive oxygen species,ROS)损伤保护作用最强。但是,法夫酵母类胡萝卜素提取物对UVA 辐照HepG2 细胞的存活率、GSH 含量和T-AOC 均无促进作用。综上,法夫酵母类胡萝卜素提取物对癌细胞ROS 氧化损伤不具有保护作用,因此可能是更有效的癌症放疗和化疗辅助治疗保护剂。
Abstract:This study aims to explore the effect of common carotenoids (carotenoids extract of Phaffia rhodozyma,astaxanthin,β-carotene,and echinenone)on HepG2 cells with oxidatively generated damage induced by ultraviolet A (UVA).Specifically,the toxicity to HepG2 cells and UVA irradiation dose were tested,and glutathione(GSH)content and total antioxidant capacity(T-AOC)were examined to clarify the effect.The results showed that astaxanthin,β-carotene,and echinenone could improve the survival rate,GSH content,and T-AOC of HepG2 cells damaged by UVA irradiation.Among them,astaxanthin demonstrated the strongest activity against the reactive oxygen species(ROS)damage to HepG2 cells.However,the carotenoid extract of P.rhodozyma failed to improve the survival rate,GSH content,and T-AOC of HepG2 cells damaged by UVA irradiation.In summary,the carotenoid extract of P.rhodozyma failed to protect cancer cells from ROS damage,and thus it is a potential protective agent in chemoradiotherapy.
