中文摘要: 为开发针对水生生物体内典型土腥味物质的快速检测方法,以2-异丁基-3-甲氧基吡嗪(2-isobutyl-3-methoxypyrazine,IBMP)为检测内标,基于分散固相萃取技术与气相色谱串联质谱联用技术开展鱼组织中土臭素(geosmin,GSM)和2-甲基异莰醇(2-methyli-soborneol,2-MIB)的检测方法开发。采用乙腈作为提取溶剂,试样提取后经脱水和吸附净化,气相分离后采用选择离子(selected ion monitor,SIM)监测模式测定。对鱼组织样品的检测结果表明,GSM 和 2-MIB 在 0.50 μg/L~500.00 μg/L 线性良好,相关系数 R2≥0.999,检出限分别为 0.37 ng/g和 0.51 ng/g,方法的定量限分别为1.23 ng/g和1.69 ng/g。3个水平的加标试验表明,方法中GSM和2-MIB的加标平均回收率为99.7%~115.7%(n=3),相对标准偏差为3.0%~14.0%,方法能够满足对不同鱼类样品的检测。

Abstract:To develop a rapid detection method for typical off-flavor;in aquatic organisms,this study used 2-isobutyl-3-methoxypyrazine(IBMP)as the internal test target and developed the detection methods of geosmin(GSM)and 2-methyli-soborneol(2-MIB)in fish tissues based on the dispersed solid phase extraction technology and gas chromatography-tandem mass spectrometry.The method used acetonitrile as the extraction solvent.After extraction,the specimen was dehydrated,adsorbed,purified,and determined by the selected ion monitor(SIM)after gas-phase separation.The test results of the fish tissue samples showed that GSM and 2-MIB were good linear in the range of 0.50 μg/L-500.00 μg/L,with correlation coefficients R2≥0.999.The limits of detection were 0.37 ng/g and 0.51 ng/g,respectively,and the limits of quantification of the methods were 1.23 ng/g and 1.69 ng/g,respectively.Three levels of standardization experiments showed that the spiked recoveries of GSM and 2-MIB in the method were 99.7%-115.7%(n=3),and the relative standard deviation(RSD)was 3.0%-14.0%.The method met the testing of different fish samples.
