中文摘要: 以金耳为原料,通过水提、醇沉、脱蛋白、透析、冷冻、干燥得到金耳多糖(Tremella aurantialba polysaccharide,TAP)。采用咔唑-硫酸比色法测定TAP糖醛酸含量,考马斯亮蓝法鉴定TAP蛋白质含量,苯酚硫酸法测定TAP多糖含量,动物实验研究TAP对环磷酰胺(cyclophosphamide,Cy)诱导免疫力低下小鼠的保护作用。结果表明,TAP糖醛酸含量为10.08%,蛋白质含量为0.7%,多糖含量为88.29%。动物实验表明TAP能增加小鼠体质量、胸腺指数,促进小鼠饮食;血细胞分析表明TAP能有效增加血细胞内白细胞、淋巴细胞、单核细胞、中性粒细胞数目;降低结、盲肠pH值;促进小肠组织分泌性免疫球蛋白A(secretory immunoglobulin A,SIgA)分泌,表明TAP能够有效逆转环磷酰胺诱导的免疫力低下小鼠的生理状况。
Abstract:Tremella aurantialba polysaccharide(TAP)was obtained by water extraction,alcohol precipitation,deproteinization,dialysis,freezing and drying.The contents of glucuronic acids,proteins and polysaccharides in TAP were determined by carbazole-sulfuric acid colorimetry,Coomassie bright blue staining and phenolsulfuric acid method,respectively.The protective effect of TAP on cyclophosphamide(Cy)-induced immunocompromised mice was investigated via animal experiments.The results showed that the contents of glucuronic acids,proteins and polysaccharides in TAP were 10.08%,0.7% and 88.29%,respectively.Animal experiments revealed that TAP increased the body weight,thymus index and daily food intake of mice.It was found in blood cell analysis that TAP elevated the number of white blood cells,including lymphocytes,monocytes and neutrophils,reduced the pH of colon and cecum,and promoted the secretion of secretory immunoglobulin A(SIgA)in small intestine,indicating that TAP effectively reversed the physiological condition of Cy-induced immunocompromised mice.
