中文摘要: 衣康酸是通过微生物发酵生产的一种有机酸,广泛应用于化工和医药领域。该文基于衣康酸和高锰酸钾发生的氧化还原反应,建立高锰酸钾分光光度法快速测定衣康酸浓度的方法。该测定方法最大检测波长540 nm,高锰酸钾(2.50 g/L)2.0 mL,样品1 mL,反应温度30 ℃,反应时间30 min。衣康酸浓度在0.12 g/L~0.36 g/L 的范围内,吸光度与浓度呈现线性关系。利用此方法测定衣康酸浓度,相关系数R2 为0.995 3,测得的相对标准偏差在±5.00%范围内,回收率在100.00%~101.11%之间,结果可信。该方法可用于快速测定发酵液中衣康酸的浓度。
Abstract:Itaconic acid is an organic acid produced by microbial fermentation,which is widely used in the chemical and pharmaceutical fields.Based on the redox reaction of itaconic acid with potassium permanganate,this paper established a method of potassium permanganate spectrophotometric rapid determination of itaconic acid.The maximum detection wavelength of this method was 540 nm,the potassium permanganate(2.50 g/L)was 2.0 mL,the sample was 1 mL,the reaction temperature was 30 ℃,and the reaction time was 30 min. The absorbance and concentration showed a linear relationship when itaconic acid concentration was in the range of 0.12 g/L-0.36 g/L.This method was used to determine the concentration of itaconic acid,the correlation coefficient R2 was 0.995 3,the measured relative standard deviation was within ±5.00%,and the recovery was 100.00%-101.11%.The results were reliable.The method established in this paper can be applied to the determination of itaconic acid in the fermentation broth.
