中文摘要: 研究酶法提取葛氏鲈塘鳢鱼头多糖的最佳条件,并探讨其体外抗氧化活性。在单因素试验的基础上,采用响应面法优化葛氏鲈塘鳢多糖提取工艺,以羟自由基和DPPH 自由基清除能力考察体外抗氧化活性并通过傅里叶变换红外光谱测定多糖的结构。试验得到最佳工艺参数:pH6.5、酶添加量2.5%、料液比1∶30(g/mL)、酶解温度37 ℃、酶解时间2 h,在此条件下多糖的提取率为11.43%。体外抗氧化试验表明,在浓度为10 mg/mL 时,多糖的羟自由基和DPPH 自由基清除率分别为81.2%和80%。红外光谱表明葛氏鲈塘鳢鱼头多糖可能是含有β-糖苷构型和吡喃环的酸性多糖。以上结果初步揭示了酶法提取葛氏鲈塘鳢鱼头多糖的特性,为水产品副产物高值化利用提供了理论基础。
Abstract:The enzyme extraction conditions of polysaccharides from the head of Perccottus glenii were optimized,and the antioxidant activity of the extracted polysaccharides in vitro was determined.On the basis of single factor tests,response surface methodology was employed to optimize the extraction process of polysaccharides from P.glenii.The antioxidant activity of polysaccharides in vitro was evaluated based on the hydroxyl and DPPH free radical scavenging rates,and the structure of polysaccharides was determined by Fourier transform infrared spectroscopy(FTIR).The extraction conditions were optimized as pH6.5,enzyme dosage of 2.5%,solid-to-liquid ratio of 1∶30(g/mL),and enzymolysis at 37 ℃for 2 h,under which the yield of polysaccharides was 11.43%.The in vitro antioxidant test showed that the hydroxyl and DPPH free radical scavenging rates of 10 mg/mL polysaccharides were 81.2% and 80%,respectively. The FTIR result showed that the polysaccharides from the head of P.glenii might contain β-glycoside configuration and pyranoid ring. The findings of this study preliminarily revealed the properties of the polysaccharides from the head of P. glenii,providing a theoretical basis for the value-added application of by-products in aquatic product processing.
