中文摘要: 采用微量板半定量法分析不同外界胁迫条件对植物乳杆菌zwq9的生物膜形成能力及其产蛋白酶活力的影响。结果表明:蛋白酶K处理能够明显破坏zwq9的生物膜,而高碘酸钠对生物膜无明显影响;在乳酸胁迫条件下,乳酸浓度0.8%以上时zwq9的生长和生物膜的形成被显著抑制(P<0.05),其产酶活力也显著降低90%以上(P<0.05);在温度胁迫条件下,30℃~40℃时zwq9的生物膜形成能力和产酶活力显著高于其他处理组(P<0.05);在盐胁迫条件下,较低浓度的NaCl(1%~4%)对zwq9的生物膜形成无显著影响(P>0.05),但却显著增强其产酶活力(P<0.05),而当NaCl浓度高于8%时,zwq9的生物膜及产酶活力均下降90%以上;相关性分析结果表明,在乳酸和盐胁迫条件下,植物乳杆菌zwq9的生物膜形成能力和产蛋白酶活力呈极显著正相关关系(P<0.01)。
Abstract:The effects of different stress systems on the biofilm formation and protease-producing activity of Lactobacillus plantarum zwq9 were analyzed by microplate semi-quantitative method.The results showed that the biofilm of L.plantarum zwq9 was significantly destroyed by proteinase K,while no significant change was found in that treated with sodium periodate.Under lactic acid stress,the growth and biofilm formation of L.plantarum zwq9 were significantly inhibited(P<0.05)when the concentration of lactic acid reached above 0.8%,and the protease-producing activity of L.plantarum zwq9 was also reduced by more than 90%(P<0.05).Under culture temperature stress,the biofilm formation and protease-producing activity of L.plantarum zwq9 at 30℃-40 ℃ were significantly higher than those of other groups(P<0.05).Under salt stress,NaCl at lower concentrations(1%-4%)had no significant effect on the biofilm formation of L.plantarum zwq9(P>0.05),but significantly enhanced its protease-producing activity (P<0.05).When the concentration of NaCl was higher than 8%,the biofilm formation and protease-producing activities of L.plantarum zwq9 both decreased by more than 90%.The correlation analysis indicated that the biofilm formation and protease-producing activity of L.plantarum zwq9 had a significant positive correlation(P<0.01)under the stress conditions of lactic acid and salt.
