中文摘要: 以云南龙陵紫皮石斛鲜条为原料,采用微波辅助-浸提法提取紫皮石斛多糖(Dendrobium devonianum Paxt.polysaccharide,DDP)。以亚硒酸钠为硒化剂,冰醋酸为催化剂对DDP进行硒化修饰获得硒化紫皮石斛多糖(Se-DDP)。通过电感耦合等离子体原子发射光谱(inductively coupled plasma atomic emission spectrometer,ICP-AES)法测定硒含量,由傅里叶变换红外光谱法对Se-DDP进行表征。以硒含量为指标,通过单因素和正交试验优化硒化反应条件,并测定Se-DDP和DDP对1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl,DPPH)、OH和O2-自由基的清除能力。结果表明,Se-DDP 的最佳制备工艺为 DDP∶亚硒酸钠=1∶2.0(g/g),反应温度 90 ℃,DDP∶冰乙酸=1∶4.0(g/mL),反应时间48 h,此时Se-DDP的硒含量为12.37 mg/g。Se-DDP的抗氧化试验结果表明,当浓度高于4 mg/mL时,Se-DDP对DPPH自由基、OH自由基和O2-自由基的清除能力均强于DDP;当浓度为8 mg/mL时,Se-DDP的抗氧化能力与VC相当。
Abstract:Yunnan Longling Dendrobium devonianum Paxt.were used as the raw material,and Dendrobium devonianum Paxt.polysaccharide(DDP)were extracted using microwave-assisted immersion and selenized with sodium selenite using glacial acetic acid as the catalyst to form Se-DDP.Selenium content was determined using inductively coupled atomic emission spectroscopy,and Se-DDP was characterized using Fourier-transform infrared spectroscopy.The reaction conditions for selenium were optimized using single factor experiment and orthogonal experiment with selenium content on the refractive index.The scavenging ability of Se-DDP towards 1,1-diphenyl-2-picrylhydrazyl(DPPH),OH,and O2-free radical was determined.The optimal preparation process conditions for Se-DDP,i.e.,DDP:sodium selenite of 1∶2.0(g/g),90 ℃ reaction temperature,DDP∶ice acetic acid of 1∶4.0(g/mL),48-h reaction time,provided Se-DDP with a selenium content of 12.37 mg/g.Antioxidant experiments revealed that Se-DDP cleared DPPH,OH,and O2-free radical capacities better than DDP,when concentrations exceeded 4 mg/mL.When the concentration was 8 mg/mL,the Se-DDP antioxidant capacity was comparable to ascorbic acid(VC).
