中文摘要: 该文对蜂王浆(royal jelly,RJ)及其粗蛋白提取物、蛋白类功能因子提取物和脂类功能因子提取物的改善睡眠作用进行研究。采用超滤透析、真空冷冻干燥、甲醇-二氯甲烷提取法等一系列方法分别提取蜂王浆粗蛋白、蛋白类功能因子和脂类功能因子。通过腹腔注射对氯苯丙氨酸构建失眠小鼠模型。经蜂王浆及其蛋白质、脂类功能因子干预后,睡眠潜伏期明显缩短,睡眠时长有延长趋势,并且能够提高小鼠血清中5-羟色胺水平。同时,脂类功能因子在中、高剂量条件下能提高小鼠血清多巴胺含量,在低剂量下能明显提高小鼠脑组织中去甲状腺肾上腺素含量。试验结果表明蜂王浆及其蛋白类、脂类功能因子提取物均能改善对氯苯丙氨酸引起的失眠症状及其相关神经递质异常表达,并且脂类功能因子的效果相对更好。
Abstract:The intervention effects of royal jelly,its crude protein fractions and its functional protein and lipid extracts on insomnia were investigated.Dialysis ultrafiltration,vacuum freeze-drying and methanoldichloromethane extraction were used to obtain the above fractions from royal jelly.The insomnia model of mice was established by intraperitoneal injection of 4-chloro-DL-phenylalanine.Oral administration of royal jelly and functional protein or lipid extracts significantly improved the sleep latency,upregulated the serum level of 5-hydroxytryptamine and prolonged the average sleep duration.In addition,the functional lipid extracts further increased the serum content of dopamine remarkably under both medium and high intervention dose,and stimulated the norepinephrine production in brain tissue under low dose.Therefore,royal jelly and its functional protein and lipid fractions could generally ameliorate PCPA-induced insomnia and the associated disorders of neurotransmitters,with the functional lipid extracts performing better improvement effects.
