中文摘要: 为探究电子束辐照对核桃青皮中醌类物质提取效果及活性的影响,以及不同萃取相(石油醚、二氯甲烷、乙酸乙酯)核桃青皮提取物的抑菌活性和抗氧化活性。通过响应面法研究最优提取条件,利用牛津杯法测定不同萃取相对蜡样芽孢杆菌、大肠杆菌、沙门氏菌、金黄色葡萄球菌的抑菌活性,通过测定DPPH·、ABTS+·和·OH清除率评价不同萃取相的抗氧化活性。结果表明:电子束辐照预处理可提升核桃青皮中胡桃醌的得率,适宜的辐照剂量为30 kGy,超声提取参数为乙醇浓度65% ,料液比1∶40(g/mL),超声时间170 min,醌类物质得率达到8.35 mg/g;青皮提取物的不同萃取相具有一定的抗氧化活性和抑菌活性,其中乙酸乙酯相萃取物的抗氧化活性最强,对DPPH·、ABTS+·、·OH的清除率分别达95.8% ,70.7% 和27.7% ,同时对大肠杆菌、金黄色葡萄球菌和沙门氏菌的抑菌活性也较强,抑菌圈直径分别达到(13.81±0.42)mm,(11.70±0.32)mm和(12.86±0.85)mm。 电子束辐照耦合超声辅助提取可增强核桃青皮醌类物质的提取效果,并提升活性物的抑菌与抗氧化活性,在天然产物提取中具有广阔的应用前景。
Abstract:The present study investigated the effects of electron beam irradiation on the extraction efficiency and activity of quinones from walnut green husk and explored the antibacterial activity and antioxidant activity of different organic extraction phases(petroleum ether phase,dichloromethane phase,and ethyl acetate phase).The optimal extraction conditions were determined by response surface methodology(RSM).Antibacterial activity was evaluated by the inhibition zone size of Bacillus cereus,Escherichia coli,Salmonella,and Staphylococcus aureus by the Oxford cup method,and antioxidant activity was evaluated by scavenging rate of DPPH·,ABTS+·,and·OH.The results showed that electron beam irradiation pretreatment could increase the yield of quinones from walnut green husk and suitable radiation dose was 30 kGy.The optimum extraction conditions were as follows:ethanol concentration of 65% ,material-to-liquid ratio of 1∶40(g/mL),and ultrasonic time of 170 min.Under above conditions,the yield of quinones reached 8.35 mg/g.The different extraction phases of the green husk extract showed certain antioxidant activity and antibacterial activity.The ethyl acetate extract possessed the strongest antioxidant activity,and the scavenging rates of DPPH·,ABTS+·,and·OH reached 95.8% ,70.7% ,and 27.7% respectively.Meanwhile,ethyl acetate extract also showed strong antibacterial activity against E.coli,S.aureus,and Salmonella,and the diameters of the inhibition zone reached(13.81±0.42)mm,(11.70±0.32)mm,and(12.86±0.85)mm respectively.Electron beam irradiation coupled with ultrasound-assisted extraction could enhance the extraction efficiency of quinones from walnut green husk,and potentiate the antibacterial and antioxidant activities of active compounds,which had broad application prospects in the extraction of natural products.
