中文摘要: 为研究菲律宾蛤仔中糖胺聚糖(glycosaminoglycans,GAGs)的结构特征,采用水酶法提取菲律宾蛤仔粗多糖,利用二乙胺基乙基纤维素-650M(diethylaminoethyl cellulose-650M,DEAE-650M)离子交换柱层析进行分离纯化后,通过紫外光谱(ultraviolet,UV)、红外光谱(infrared,IR)和核磁共振(nuclear magnetic resonance,NMR)等方法对纯化后的菲律宾蛤仔GAG-1进行结构表征与分析。结果表明,采用水酶法提取的菲律宾蛤仔GAGs粗品,得率为1.04%。应用DEAE-650M离子交换柱层析分离纯化得到峰GAG-1和峰GAG-2。其中GAG-1的含量为89.56%,比粗品提高了82.43%,而GAG-2含量很低。紫外光谱分析结果表明,菲律宾蛤仔GAG-1中含微量核酸或蛋白质杂质;红外光谱分析表明GAG-1含有羧基、乙酰氨基、硫酸基等基团,其单体为吡喃型;1H-NMR和13C-NMR图谱分析结果显示GAG-1 以 β-D-Glc(1→4)-为主要连接方式。
Abstract:To study the structural characteristics of glycosaminoglycans(GAGs)from Ruditapes philippinarum,crude polysaccharides were extracted by aqueous enzymatic method.GAG-1 was isolated and purified by diethylaminoethyl cellulose-650M(DEAE-650M)ion exchange chromatography.The structure of GAG-1 was characterized by ultraviolet(UV)spectrum,infrared(IR)spectrum and nuclear magnetic resonance(NMR).The results showed that the yield of crude GAGs extracted by aqueous enzymatic method was 1.04%.For GAG-1 and GAG-2 obtained by DEAE-650M ion exchange chromatography,the content of GAG-1 was 89.56%,82.43% higher than that of crude GAGs,while the content of GAG-2 was very low.The UV spectrum revealed that GAG-1 contained trace nucleic acid or protein impurities.The IR spectrum demonstrated that GAG-1 contained carboxyl,acetyl amino,and sulfate groups,with pyran monomer.From the1H-NMR and13C-NMR analysis,it found that GAG-1 was mainly connected by β-D-Glc(1→4)-.
