生菜多酚氧化酶的酶活特性及碳点对其抑制作用

中文摘要: 多酚氧化酶(polyphenol oxidase,PPO)作为一种能催化邻苯二酚氧化成邻苯二醌的金属蛋白酶,是许多蔬菜发生酶促褐变的重要原因。研究采用分光光度法测定以不同酚(邻苯二酚、间苯二酚、对苯二酚、苯酚、邻苯三酚)为底物,在生菜PPO作用下的产物在420 nm处的吸光度变化以及温度、pH值对生菜PPO活性的影响,结果表明,生菜PPO的最适温度为40℃,最适pH值为6,该酶催化邻苯二酚的酶促反应速度最快。同时选择硫掺杂的碳点(S doped carbon,S-CDs)、铜掺杂的碳点(Cu doped carbon,Cu-CDs)、溴掺杂的碳点(Br doped carbon,Br-CDs)为研究对象,以常用的柠檬酸、抗坏血酸、半胱氨酸作对比,探讨碳点作为一种新型PPO抑制剂的可行性。试验结果表明,S-CDs表面所带的还原性基团及S原子的还原性对S-CDs的抑制效果有很大帮助,这明显区别于其它CDs和常规抑制剂。抑制剂对该酶的抑制作用大小顺序为S-CDs>抗坏血酸>柠檬酸>Cu-CDs,故可将碳点用作生菜酶促褐变的抑制剂。

Abstract:Polyphenol oxidase(PPO),a metalloproteinase which catalyzes the oxidation of o-benzenediol to o-benzodioxanone,is the main cause of enzymatic browning in many vegetables.Spectrophotometry was used to detect the absorbance of the products of different phenols(o-benzenediol,m-benzenediol,p-benzenediol,phenol,pyrogallol)under the catalysis of PPO from lettuce(Lactuca sativa L.)at 420 nm and the effect of temperature and pH on PPO activity.The result showed that the enzyme catalyzed the fastest enzymatic reaction with o-benzenediol as substrate,with the optimum temperature of 40 ℃ and optimal pH of 6.Meanwhile,Cu-doped carbon dots(Cu-CDs),Br-doped carbon dots(Br-CDs),and S-doped carbon dots(S-CDs)were selected to explore the potential of CD as a novel PPO inhibitor with common inhibitors(citric acid,ascorbic acid,and cysteine)as references.The result verified that the inhibition of S-CDs was ascribed to the reducing groups on the surface of S-CDs and the S atom.The efficiency of inhibitors on PPO was in the order of S-CDs>ascorbic acid>citric acid>Cu-CDs.
