灵芝醇提物对改善3T3-L1脂肪细胞胰岛素抵抗的作用

中文摘要: 研究讨论灵芝醇提物对地塞米松诱导的3T3-L1脂肪细胞胰岛素抵抗的影响。采用地塞米松诱导脂肪细胞胰岛素抵抗模型,将细胞分成正常组、模型组、灵芝醇提物组、二甲双胍(阳性对照)组,检测各组的葡萄糖消耗量和甘油三酯含量,分析灵芝醇提物对胰岛素抵抗的影响。建立模型的地塞米松处理组与正常组相比,1 μmoL/L地塞米松处理72 h显著降低了3T3-L1脂肪细胞的葡萄糖消耗量(P<0.05),且胰岛素抵抗状态在24 h内维持稳定,脂肪细胞胰岛素抵抗模型构建成功。10、50、100 mg/L灵芝醇提物对细胞活力均无显著影响,灵芝醇提物处理细胞24 h能显著提高脂肪细胞的葡萄糖消耗量和细胞内甘油三酯的含量且呈剂量依赖性。同时灵芝醇提物处理后显著增加了胰岛素受体底物1、葡萄糖转运体-4、磷脂酰肌醇-3-激酶p110亚基、磷脂酰肌醇-3-激酶p85亚基、磷酸化蛋白激酶、磷酸化糖原合酶激酶3β蛋白的表达。结果表明灵芝醇提物通过IRS1/PI3K/AKT信号途径改善地塞米松诱导的脂肪细胞胰岛素抵抗。
Abstract:This study investigated the effect of ethanol extract of Ganoderma lucidum on insulin resistance in 3T3-L1 adipocytes induced by dexamethasone.An adipocyte insulin resistance model induced by dexamethasone was adopted.Cells were divided into a normal group,a model group,a G.lucidum ethanol extract group,and a metformin(positive control)group.The effect of G.lucidum ethanol extract on insulin resistance was analyzed through detecting the glucose consumption and triglyceride content of each group.Compared with the normal group,1 μmol/L dexamethasone significantly reduced glucose consumption of 3T3-L1 adipocytes after treatment for 72 h(P<0.05),and the state of insulin resistance was stable within 24 h,indicating that the adipocyte insulin resistance model was successfully established.G.lucidum ethanol extract with 10,50 mg/L,and 100 mg/L had no significant effect on cell viability.G.lucidum ethanol extract significantly increased glucose consumption and intracellular triglyceride content of adipocytes in a dose-dependent manner after treatment for 24 h.At the same time,the protein expressions of insulin receptor substrate(IRS1),glucose transporter 4(GLUT4),phosphoinositide 3 kinase p110 (PI3K p110),PI3K p85,phosphor-kinase-protein kinase B (p-AKT),and phosphorylated glycogen synthase kinase-3β(p-GSK3β)was significantly increased after G.lucidum ethanol extract treatment.The results showed that G.lucidum ethanol extract ameliorated dexamethasone-induced insulin resistance in adipocytes through the expression of IRS1/PI3K/AKT signaling pathway.
