中文摘要: 为了研究枣渣中功能性物质,提高废弃物利用率,该研究采用单因素试验和响应面法相结合优化枣渣中总三萜含量(total triterpene content,TTC)的提取工艺,以常规振荡为对照,比较两种提取物的抗氧化活性及对A549癌细胞的抗增殖作用。结果表明,超声辅助最佳提取条件为超声温度60.00℃,超声时间42 min,液固比34∶1(mL/g),乙醇浓度64%,该条件下总三萜含量为15.854 mg/g。发现超声辅助提取物的抗氧化和抗增殖能力均高于振荡提取,两种枣渣总三萜提取物对DPPH、ABTS+和羟基自由基具有较好的清除作用,清除效果与总三萜浓度呈剂量-效应关系;当提取物浓度为1 mg/mL时,两种提取物抗A549细胞增殖效果最好,其中超声辅助提取物为37.62%,常规振荡提取物为27.18%。枣渣中三萜具有良好的抗氧化活性和抗增殖能力。
Abstract:This paper aims to explore the functional substances in jujube residue and improve the utilization rate of waste.To be specific,the ultrasonic-assisted extraction(UAE)of total triterpenoids in jujube residue was optimized by single factor test and response surface methodology(RSM),with the conventional extraction by oscillation(COE)as the control.The antioxidant activity and the anti-A549 cell proliferation activity of the two extracts were compared.The results showed that the optimal UAE conditions were as follows:ultrasonic temperature of 60 ℃,ultrasonic time of 42 min,liquid-to-solid ratio of 34∶1(mL/g),and 64% ethanol.Under the above conditions,the total triterpenoid content(TTC)was 15.854 mg/g.The antioxidant and anti-proliferation activity of the ultrasonic-assisted extract was both higher than that of the extract yielded by COE.The two triterpenoid extracts of the jujube residue displayed strong DPPH radical-,ABTS+radical-,and hydroxyl radical-scavenging ability,and the ability was dose-dependent.At the concentration of 1 mg/mL,the extracts yielded by UAE and COE showed the strongest anti-proliferation activity,and the cancer cell-inhibiting rate reached 37.62% and 27.18%,respectively.Triterpenoids from jujube residue have strong antioxidant activity and anti-proliferation ability.
