中文摘要: 为确定从牡丹籽粕中提取牡丹籽多糖的工艺参数,以水浴温度、超声时间、料液比、提取次数为因素,在单因素试验的基础上,采用响应面法优化超声波辅助热水浸提多糖的最佳工艺,并通过DPPH自由基和ABTS+自由基清除能力对牡丹籽多糖的抗氧化活性进行初步研究。结果表明:牡丹籽多糖的最佳提取工艺为水浴温度85℃、超声时间40 min、料液比1∶60(g/mL)、提取次数2次。该条件下提取牡丹籽多糖得率为(44.48±1.26)%;抗氧化活性试验结果显示,牡丹籽多糖对ABTS+自由基和DPPH自由基的清除能力均随多糖浓度的增大而增大,在多糖浓度为4 mg/mL时,牡丹籽多糖对ABTS+自由基和DPPH自由基清除率分别为(40.39±1.22)%和(38.38±1.17)%。
Abstract:To determine the extraction process parameters of polysaccharides in peony seed meal(PSP),the extraction process of polysaccharides,with ultrasonic-assisted hot water extraction,was optimized by response surface methodology (RSM)based on single factor test,with water bath temperature,ultrasonic time,solidliquid ratio,and extraction times as the factors.The antioxidant activity of polysaccharides from peony seed was preliminarily evaluated based on DPPH radical and ABTS+radical scavenging ability.Results showed that the optimal extraction conditions were as follows:water bath temperature,85℃;ultrasonic time,40 min;solidliquid ratio,1 ∶60 (g/mL);and extraction times,2 times.Under these conditions,the extraction yield of PSP was (44.48±1.26)%.The results of antioxidant activity showed that the scavenging ability of ABTS+radical and DPPH free radicals increased with increase in polysaccharide concentration.When the concentration of PSP was 4 mg/mL,the scavenging rates of ABTS+radical and DPPH free radicals were (40.39±1.22)% and (38.38±1.17)%,respectively.
