低共熔溶剂提取灵芝多糖的工艺优化及抗氧化活性研究

中文摘要: 采用5种低共熔溶剂(deep eutectic solvents,DESs)提取灵芝多糖(Ganoderma lucidum polysaccharide,GLP),以灵芝多糖得率为指标筛选出最适合的低共熔溶剂是DESs-5即氯化胆碱-尿素(choline chloride-Urea,ChCl-Urea),然后利用响应面法优化灵芝多糖的提取工艺。结果表明,最佳提取条件为DESs-5含量34%,提取时间1.9 h,醇沉时间7.3 h,灵芝多糖得率可达到1.10%。与常规热水浸提法获得的灵芝多糖(Ganoderma lucidum polysaccharide-W,GLP-W)相比,采用该方法获得的灵芝多糖(Ganoderma lucidum polysaccharide choline chloride-Urea,GLP-5)得率提高了83.33%,DPPH自由基清除率提高了20.50%,还原力提高了47.47%,因此DESs作为新型绿色试剂其提取的灵芝多糖不仅得率高而且还具有较强的抗氧化活性。
Abstract:Five kinds of deep eutectic solvents (DESs) were chosen to extract Ganoderma lucidum polysaccharide(GLP),choline chloride-Urea(ChCl-Urea)(DESs-5)was regarded as the most suitable DESs according extraction yield of GLP.Then response surface method was used to optimize the extraction process of GLP.The results showed that the optimal extraction condition was 34% the content of DESs-5,1.9 h extraction time,7.3 h ethanol precipitation time,and the yield of GLP could reach 1.10%.Compared with Ganoderma lucidum polysaccharide-W(GLP-W)which obtained by the conventional water extraction technology,the extraction yield,DPPH radical scavenging rate and reducing power capacity of GLP-5 were increased by 83.33%,20.50% and 47.47%,respectively.As a kind of new green reagent,DESs can not only improve the yield of GLP,but also have notable antioxidant activity.
