中文摘要: 建立高效液相色谱(high performance liquid chromatography,HPLC)法测定核桃仁中鞣花酸含量,对比分析不同品种及温185核桃仁不同部位中的鞣花酸含量。确定检测鞣花酸的色谱条件为Phenomenex Synergi Hydro-RP C18色谱柱(4.6 mm×250 mm,4 μm)梯度洗脱,检测波长254 nm,柱温:35℃,流速1 mL/min。使用该方法测得山西古县辽河1号、古县绵核桃仁鞣花酸含量分别是(99.26±1.06)、(90.53±4.31)mg/100 g,新疆温185、新2核桃仁鞣花酸含量分别是(51.39±1.72)、(76.46±7.55)mg/100 g;新疆温185核桃仁中约60%鞣花酸来源于种皮,其含量高达(585.93±31.50)mg/100 g种皮,40%来源于脱皮仁,其含量仅为(20.52±0.38)mg/100 g脱皮仁。
Abstract:A high performance liquid chromatography (HPLC)method was established to quantitate ellagic acid in walnut kernels.Ellagic acid content in different walnut varieties,and different Wen 185 walnut kernel tissues was analyzed,and values were compared.Separations were performed on a Phenomenex Synergi Hydro-RP C18 column(4.6 mm×250 mm,4 μm)with gradient elution.The detection wavelength was 254 nm,column temperature was 35℃,and flow rate was 1mL/min.By this method,the respective ellagic acid contents of Liaohe 1 and Guxian Mian walnuts from Guxian County,Shanxi Province were(99.26±1.06)mg/100 g and(90.53±4.31)mg/100 g.The respective ellagic acid contents of Wen 185 and Xin 2 walnuts from Xinjiang Province were (51.39±1.72)mg/100 g and (76.46±7.55)mg/100 g.About sixty percent of the ellagic acid in Wen 185 walnut kernels came from the seed coat,with seed coat content reaching (585.93±31.50)mg/100 g seed coat.The remaining 40% came from peeled walnut kernel,whose content was only(20.52±0.38)mg/100 g peeled walnut kernel.
