中文摘要: 建立高效液相色谱技术检测鸡蛋中牛磺酸含量的方法,并对市售A、B品牌乌鸡蛋,C、D品牌柴鸡蛋以及普通鸡蛋蛋黄中的牛磺酸含量进行分析比较。试验采用邻苯二甲醛(ortho-phthalaldehyde,OPA)为衍生剂,利用自动进样器的在线衍生功能进行柱前衍生,流动相为甲醇+乙腈∶水(25∶75,体积比),荧光法定量。结果表明:使用该方法,牛磺酸在 1 μg/mL~25 μg/mL 具有良好的线性关系(R2=0.999 9),检测限为 0.01 μg/mL,回收率范围为 99.7%~100.5%,相对标准偏差(relative standard deviation,RSD)为0.5%~0.6%;用该方法测定A、B品牌乌鸡蛋,C、D品牌柴鸡蛋和普通鸡蛋的蛋黄,牛磺酸含量分别为1.332、1.151、1.063、0.735、0.622 mg/100 g,普通鸡蛋蛋黄中的牛磺酸含量显著低于A、B品牌乌鸡蛋和C品牌柴鸡蛋(P<0.05),略低于D品牌柴鸡蛋(P>0.05)。该方法简单易行,回收率高,精密度高,特异性强,适用于测定鸡蛋中牛磺酸含量。
Abstract:A method for the determination of taurine content in eggs by high performance liquid chromatography(HPLC)was established,and the taurine contents in egg yolks of dark eggs(brands A and B),native eggs(brands C and D),and ordinary eggs was analyzed and compared.In the experiment,ortho-phthalaldehyde was used as the derivating agent,and the on-line derivatization function of the auto sampler was used for precolumn derivatization.A methanol/acetonitrile:water (25∶75,volume ratio)mobile phase and fluorescence detection were used.The results demonstrated good linearity in the concentration range of 1 μg/mL to 25 μg/mL(R2=0.999 9).The limit of detection was 0.01 μg/mL,the recovery range was 99.7% to 100.5%,and the relative standard deviation (RSD)range was 0.5% to 0.6%.The taurine content of brands A and B (dark eggs),brands C and D (native eggs),and ordinary eggs was 1.332,1.151,1.063,0.735,0.622 mg/100 g,respectively.The taurine content in the yolk of ordinary eggs was significantly lower than that in brands A and B(dark eggs)and brand C(native eggs,P<0.05),and slightly lower than that in brand D(native eggs,P>0.05).The method was simple,accurate,specific,and suitable for the determination of taurine content in eggs.
