巨峰葡萄籽原花青素的纯化与抗氧化活性分析

中文摘要: 该文通过对影响大孔树脂HPD-400吸附、洗脱因素进行正交优化与验证,优化巨峰葡萄籽原花青素纯化工艺,并以赤霞珠葡萄籽作为参照,对其进行抗氧化活性分析。选取吸附时间、提取液pH值、上样流速、乙醇浓度、洗脱液pH值5个因素,分别考察其对原花青素浓度及吸附率、洗脱率的影响。选择提取液pH值、上样流速、乙醇浓度、洗脱液pH值进行正交试验。通过正交试验得到最优纯化工艺为提取液pH 5.0,上样流速1.0 mL/min,洗脱液pH 7.5、乙醇浓度50%,获得的原花青素纯度为94.17%。DPPH自由基清除能力、还原性强弱顺序依次为赤霞珠>巨峰>VC;超氧阴离子自由基清除能力强弱顺序依次为VC>赤霞珠>巨峰。纯化后巨峰葡萄籽原花青素抗氧化活性较好,表现出较大的应用潜力。
Abstract:The purification process of proanthocyanidins from Jufeng grape seeds was optimized by orthogonal optimization and verification of factors affecting adsorption and elution of macroporous resin HPD-400,and its antioxidant activity was analyzed with Cabernet Sauvignon seeds as reference.Five factors,including adsorption time,pH value of extraction solution,loading flow rate,ethanol concentration and pH value of elution solution,were selected to investigate the effects of these factors on procyanidins concentration,adsorption rate and elution rate.The pH value of extraction solution,loading velocity,ethanol concentration and eluent pH value were selected for orthogonal test.The optimal purification process was obtained by orthogonal test as follows:pH 5.0,loading flow rate 1.0 mL/min,pH 7.5,ethanol concentration 50%,purity of procyanidins 94.17%.DPPH free radical scavenging ability and reducing ability were Cabernet Sauvignon> Jufeng>VC.The scavenging ability of superoxide anion radical was VC>Cabernet Sauvignon> Jufeng.The purified proanthocyanidins of Jufeng grape seed showed good antioxidant activity and showed great application potential.
