中文摘要: 利用超声辅助提取法对桃金娘叶三萜进行提取,采用单因素试验和响应面法优化超声辅助提取工艺,通过香草醛-高氯酸法对三萜含量进行测定,并探讨其抗氧化能力。结果表明,各因素对三萜含量的影响大小依次为超声功率>液料比>超声时间,三萜最佳提取工艺为提取溶剂乙酸乙酯,液料比50∶1(mL/g)、超声功率294 W,超声时间90 min。在最优的提取条件下,桃金娘叶三萜的平均含量为15.72 mg/g,优于传统浸提法和水浴提取法。在桃金娘叶三萜浓度为12.27 μg/mL时,对DPPH自由基清除率可达92.52%,具有较强的抗氧化性。
Abstract:The triterpenoids of Rhodomyrtus tomentosa leaves were extracted by ultrasonic assist extraction.The process of extracting triterpenoids from Rhodomyrtus tomentosa leaves was optimized by single factor and response surface methods.The content of triterpenoids was determined by a vanillin-perchloric acid method and its antioxidant capacity was analyzed.The results showed that the effects of various factors on the extraction content of triterpenoids were in the following order:ultrasonic power>liquid material ratio>ultrasonic time.The optimum extraction conditions of triterpenoids were as follows:ethyl acetate as extraction solvent,liquid material ratio 50∶1 (mL/g),ultrasonic power 294 W and ultrasonic time 90 min.Under the optimal conditions,the content of triterpenoids in Rhodomyrtus tomentosa leaves was 15.72 mg/g,which was better than that obtained using traditional extraction and water bath extraction methods.When the concentration of triterpenoids reached 12.27 μg/mL,the DPPH free radical scavenging rate reached a maximum of 92.52%,indicating an excellent free radical-scavenging activity in vitro.
