金线莲多糖纯化工艺及其抗氧化活性研究

中文摘要: 以金线莲多糖粗提取物为原料,通过静态吸附-解吸试验,考察8种不同极性大孔树脂对多糖吸附率、解吸率、脱色率和脱蛋白率的影响,筛选出最优大孔树脂;以吸附、解吸效果为指标,考察大孔树脂纯化多糖的工艺参数;以维生素C(vitamin C,VC)为对照品,通过测定1,1-二苯基-2-三硝基苯肼自由基(DPPH·)和羟基自由基(·OH)清除能力来评价纯化前后金线莲多糖的抗氧化活性。结果表明:最优大孔树脂为X-5,金线莲多糖的最佳纯化工艺:上样浓度为4 mg/mL,上样流速为2 BV/h,上样体积为4 BV,洗脱剂浓度为40%乙醇溶液,洗脱流速为2 BV/h,洗脱量为5 BV。在此工艺下,金线莲多糖由深褐色变为浅棕色,含量从27.88%提高到76.07%;纯化后的金线莲多糖对DPPH·和·OH具有更强的清除作用,当浓度为0.6 mg/mL时,对DPPH·和·OH的清除率分别从57.27%和60.12%提高至86.24%和88.63%,清除作用略低于VC,表明纯化后的金线莲多糖具有较好的抗氧化活性,可为金线莲多糖的工业化生产和进一步开发利用提供参考。
Abstract:Eight kinds of macroporous resins were screened by adsorption efficiency,desorption efficiency,decolorization efficiency,and deproteinization efficiency of static adsorption,as well as desorption test with extraction of polysaccharides from Anoectochilus roxburghii.Using the adsorption and desorption results as indicators,the optimum macroporous resin was selected to investigate the technological parameters of purified polysaccharides.With vitamin C(VC)as the reference substance,the antioxidant ability of polysaccharides from Anoectochilus roxburghii before and after being purified was evaluated by determining the scavenging capacity of 1,1-dipheny1-2-picryl-hydrazyl hydrazine free radical(DPPH·)and hydroxyl free radical(·OH).Results showed that the optimum macroporous resin was X-5.By a single factor test,the optimal sample loading and elution conditions were as follows:4 mg/mL of polysaccharides were loaded on the resin column with a volume of 4 BV at a flow rate of 2 BV/h and eluted with 5 BV of 40% ethanol at a flow rate of 2 BV/h.Under optimum conditions,the total polysaccharide content in Anoectochilus roxburghii increased from 27.88% to 76.07%,and the color of polysaccharides changed from brunette to light.The purified polysaccharides had a strong scavenging effect on DPPH·and·OH;when the polysaccharide concentration was 0.6 mg/mL,the clearance rate of DPPH·and hydroxyl radicals increased from 57.27% to 86.24% and 60.12% to 88.63%,respectively.Although it was lower than the scavenging effect of VC,the purified polysaccharides had an acceptable antioxidant ability,providing a reference for the industrial production and further development of polysaccharides from Anoectochilus roxburghii.
