中文摘要: 以鸡血血红蛋白为原料,采用中性蛋白酶酶解,以水解度为指标确定血肽酶解制备条件,应用高效液相色谱分析血肽相对分子质量分布,通过交联葡聚糖凝胶Sephadex G-15对血肽进行分离纯化,分别测定血肽粗品、纯化后组分ABTS+自由基、DPPH自由基、羟基自由基与O2-自由基清除率。血肽酶解条件为血红蛋白底物浓度5%、酶添加量1.5%、酶解时间5 h,浓缩冻干得到血肽粗品;血肽相对分子质量主要分布在500 Da~1 000 Da与180 Da~500 Da,分别占比23.97%、66.07%;血肽经Sephadex G-15层析主要分离出4个洗脱峰,与高效液相色谱分析结果一致;180 Da~500 Da血肽表现出更高的抗氧化性,ABTS+自由基、DPPH自由基、OH自由基与O2-自由基清除率分别为81.28%、52.45%、23.86%与18.14%。

Abstract:Hemoglobin of chicken blood was hydrolyzed by neutral protease to obtain hemepeptides,and the enzymolysis conditions were determined by the degree of hydrolysis.The relative molecular mass distribution of hemepeptides were analyzed by high performance liquid chromatography.The hemepeptides were separated and purified by Sephadex G-15.Scavenging rates of ABTS+free radicals,DPPH free radicals,OH free radicals and O2-free radicals were also determined for both the crude sample and purified components.The results indicated that the optimal enzymatic parameters for preparing hemepeptides were determined as follows that substrate concentration 5.0%,enzyme concentration 1.5%,and enzymolysis time 5 h.The relative molecular mass distributions of hemepeptides were 500 Da-1 000 Da and 180 Da-500 Da,and content proportions were 23.97%and 66.07%respectively.Four main elution peaks were separated by Sephadex G-15 chromatography,consistent with high performance liquid chromatography(HPLC)analysis.The hemepeptide of 180 Da-500 Da showed stronger radical-scavenging ability,and the scavenging rates of ABTS +,DPPH,OH and O2-radicals were 81.28%,52.45%,23.86% and 18.14%,respectively.
