三产地百香果叶总黄酮提取工艺优化及其抗氧化性研究

中文摘要: 以广西、福建和四川三产地百香果叶为研究材料,运用超声辅助乙醇提取法制备百香果叶总黄酮,在单因素试验基础上,以芦丁为标准品建立总黄酮含量测定方法,将总黄酮含量作为考察指标,采用正交试验优化百香果叶总黄酮提取工艺,并评价三产地百香果叶体外抗氧化活性。结果表明:百香果叶总黄酮最优提取工艺为液料比70∶1(mL/g),超声时间30 min,乙醇含量40%,在此提取条件下,广西北流百香果叶总黄酮含量为11.55 mg RT/g DW,福建南平百香果叶总黄酮含量为13.67 mg RT/g DW,四川凉山百香果叶总黄酮含量为14.86 mg RT/g DW,体外抗氧化活性以四川凉山样品最优。

Abstract:Ultrasonication-assisted ethanol extraction of total flavonoids from Passiflora edulis Sims leaves was optimized using an orthogonal experimental design.Total flavonoids from leaves of P.edulis Sims grown in Guangxi,Fujian,and Sichuan were determined using an AlCl3 colorimetric method with rutin as a standard,and their antioxidant activities were also evaluated.The results showed that the optimal extraction conditions of total flavonoids from leaves of P.edulis Sims included a liquid∶material ratio of 70∶1(mL/g),ultrasonication for 30 min,and 40%ethanol.Under these conditions,the total flavonoids content of leaves of P.edulis Sims from Guangxi Beiliu was 11.55 mg RT/g DW,those of leaves from Fujian Nanping was 13.67 mg RT/g DW,and of leaves from Sichuan Liangshan was 14.86 mg RT/g DW.In-vitro antioxidant activity of total flavonoids were highest in the sample from Sichuan Liangshan.
