中文摘要: 为确定盐地碱蓬粗蛋白提取的最佳工艺参数,并检测其功能特性及体外抗氧化活性,采用盐析法对盐地碱蓬中的粗蛋白进行提取,通过单因素试验与响应面优化,得到最佳工艺参数;以冻干后的粗蛋白为样品,大豆蛋白为对照品,测定其在不同pH值下的功能特性;分别以VC、VE为对照品进行体外抗氧化活性分析。结果表明:盐地碱蓬粗蛋白提取最佳工艺参数为磷酸盐缓冲液(phosphate buffered solution,PBS)浓度0.04 mol/L,超声时间30 min,料液比1∶13(g/mL),静置时间61 min,粗蛋白提取率为(47.03±0.34)%;功能特性试验表明盐地碱蓬粗蛋白具有良好的持油性、溶解度、乳化性和起泡性。体外抗氧化活性试验表明,盐地碱蓬粗蛋白对1,1-二苯基-2-三硝基苯肼(1,1-dipheny1-2-picrylhydrazyl,DPPH)自由基具有一定的清除作用、对磷钼络合物具有一定的还原能力。
Abstract:In this study,optimal process parameters for the extraction of crude protein from Suaeda salsa using a salting out method were determined,and the functional properties and in vitro antioxidant activities of the extracted protein were evaluated.Freeze-dried crude protein was used as the sample material and soy protein as a reference substance to determine functional properties at different pH values,whereas VCand VEwere used as reference substances for in vitro antioxidant activity analyses.The best process parameters were determined based on single-factor analysis and response surface optimization.The results revealed that the best process parameters for extracting crude protein from S.salsa were a phosphate-buffered solution concentration of 0.04 mol/L,ultrasonic disruption for 30 min,a material-to-liquid ratio of 1∶13(g/mL),and a standing time of 61 min.Using these parameters,a protein extraction rate of 47.03% ±0.34% was obtained.Functional characterization indicated that the extracted S.salsa crude protein had good oil retention,solubility,emulsification,and foaming properties,and in vitro antioxidant analyses revealed that the protein had a certain DPPH radical scavenging effect,as well as the capacity to reduce phosphomolybdenum complexes.
