中文摘要: 为开发利用浒苔多糖,制备高抗氧化活性的低聚糖水解物,以条浒苔为材料,采用纤维素酶辅助碱提法提取浒苔粗多糖,通过酸水解法将多糖降解为低聚糖。以羟自由基清除率为指标,采取单因素试验,研究不同料液比、酸浓度、时间、温度对浒苔多糖酸水解的影响,确定影响酸水解的关键因素及其适宜参数范围,然后采用响应面分析法优化水解工艺。结果表明:多糖酸水解最佳条件为料液比1∶40(g/mL)、温度为70 ℃、盐酸浓度1.4 mol/L、时间2 h,在此条件下羟自由基清除率模型预测值为80.40%,实测值为(79.76±0.82)%。

Abstract:The aim of this study was to prepare oligosaccharides with high antioxidant activity using polysaccharides derived from Enteromorpha prolifera.Polysaccharides from E.prolifera were extracted through the cellulase-assisted extraction method,which were then degraded into oligosaccharides by acid hydrolysis. The hydroxyl radical scavenging rate were used as index.The single-factor test was used to study the effects of material/water ratio,acid concentration,time,and temperature on the acid hydrolysis of polysaccharide to identify the key factors and appropriate parameters.Further,the response surface analysis methodology was used to optimize the hydrolysis process conditions.The results showed that the optimum conditions for acid hydrolysis of polysaccharides were as follows: 1∶40 (g/mL)material/water ratio,70 °C temperature,and 1.4 mol/L of hydrochloric acid for 2 h. Under these conditions, the model predictive value of hydroxyl radical scavenging activity was 80.40%,and the measured value was(79.76±0.82)%.
