中文摘要: 该文旨在探究核桃低聚肽(walnut oligopeptides,WOPs)对吲哚美辛诱导的大鼠胃溃疡的保护作用。大鼠被随机分为8 组(n=10):正常对照组、模型对照组、乳清蛋白对照组(440 mg/kg bw)、奥美拉唑对照组(20 mg/kg bw)、3 个WOPs 剂量组(220、440、880 mg/kg bw)及1 个与牛骨胶原低聚肽(bovine collagen oligopeptides,BCOPs)的配伍组(WOPs 440 mg/kg bw+BCOPs 1.5 g/kg bw)。灌胃干预30 d 后,采用吲哚美辛48 mg/kg 腹腔注射造模,4 h 后,快速分离胃组织,进行胃黏膜损伤观察及评分,检测全血血常规及胃组织前列腺素E2(prostaglandin E2,PGE2)、一氧化氮(nitric oxide,NO)、髓过氧化物酶(myeloperoxidase,MPO)的水平。结果发现WOPs 干预明显降低了胃黏膜损伤积分指数,提高了PGE2 合成水平,抑制了MPO 和NO 水平的升高。表明WOPs 对吲哚美辛所诱导的大鼠实验性胃溃疡具有较好的保护作用,其主要作用机制与减轻侵袭因素对胃黏膜的直接损伤、增加胃组织PGE2 含量、抑制中性粒细胞的聚集浸润及NO 的水平有关。
Abstract:The study investigated the protective effect of walnut oligopeptides(WOPs)against indomethacininduced gastric ulcer in rats. Rats were randomly divided into eight groups(n = 10),normal group,ethanol group,whey protein group(440 mg/kg bw),omeprazole group(20 mg/kg kw),three WOPs groups(220, 440,880 mg/kg bw, respectively)and one combination group(WOPs 440 mg/kg bw + BCOPs 1.5g/kg bw)with bovine collagen oligopeptides(BCOPs).After oral gavage once a day for 30 consecutive days,rats were injected intraperitoneally 48 mg/kg indomethacin to induce gastric ulcer.After four hours,gastric tissue was rapidly separated,then the gross lesions of gastric tissue was observed and evaluated,the whole blood routine and gastric prostaglandin E2(PGE2),nitric oxide(NO)and myeloperoxidase(MPO)levels were detected. The results showed that the administration with WOPs reduced indomethacin-induced punctate or banded gastric mucosal injury,enhanced the synthesis of PGE2 ,while suppressed the elevated of PGE2 and NO levles. These results suggested that WOPs have protective effects against indomethacin-induced gastric ulcer in rats. Its gastroprotective effects may be possibly assign to its effect on attenuating the severity of gastric mucosal damage caused by aggressive factors,enhancing PGE2,suppressing neutrophils infiltration and elevation of NO levels.
