响应面法优化荔枝壳总黄酮提取及抗氧化活性

中文摘要: 以荔枝壳为原料,采用乙醇加热回流提取荔枝壳中的总黄酮。研究提取工艺条件对荔枝壳总黄酮得率的影响后,进一步通过响应面法对工艺条件参数进行优化。结果表明,回归模型具有高度显著性,最佳提取工艺条件为料液比1∶19(g/mL),浸提温度65℃,浸提时间127 min,乙醇体积分数67%。在最佳工艺条件下,总黄酮得率预测值为7.07%,验证值为7.03%,与模型预测值相对误差仅为0.57%。荔枝壳总黄酮提取液在测定浓度范围内对DPPH·、NO2-和O2-·有较好的清除能力,同时还具有较强的还原力。

Abstract:Base on a single factor experiment,a quadratic regression model was established.Thereafter,the extraction process of total flavonoids from litchi shells was optimized using the response surface methodology.The results showed that the quadratic regression model exhibited high significance,and the optimal values of the variables were as follows:solid to solvent ratio of 1∶19 (g/mL),extraction temperature of 65 ℃,extraction time of 127 min and ethanol concentration in aqueous solution of 67%.Under these conditions,the predicted and actual yields of total flavonoids were 7.07% and 7.03%,respectively,and the relative error was only 0.57%.The total flavonoids extracted from litchi shells exhibit a good scavenging ability for DPPH·,NO2-and O2-·within the measured concentration range,along with a strong reducing power.
